| ObjectiveAsthma is a variety of cells, especially mast cells, eosinophils, T lymphocytes, neutrophils, airway epithelial cells and a variety of cellular components involved in the chronic airway inflammation and disease, immune regulation plays an important role in asthma pathogenesis, Mechanism of selenomethionine in immunology are taken more attention,To investigate the effects of selenomethionine on interleukin-4(IL-4) and interferon-γ(interferon-γ, IFN-γ) in bronchoalveolar lavage fluid(BALF), Cell count and classification,and expression of intercellular adhesion molecular-1(ICAM-1) in the lung tissue in asthmatic rats.MethodsIn accordance with the principles of randomization, 50 male SD rats were randomly divided into five groups:asthma model group, dexamethasone treatment group, normal control group, selenomethionine low dose group and high- dose group;(1) Asthmatic group: Rats were immunized on day 1, day 8 and day 15 by intraperitoneal injection of 100 mg ovalbumin(OVA) in 1 ml of saline with 100 mg of aluminum hydroxide. From the sixteenth day rats will have been sensitized on the self-made atomization box,with 1% ovalbumin atomization excitation, 30 minutes a day, for 7 consecutive days. Asthma rats as easily angered shortness of nose,incontinence and so on.OVA sensitized rats were injected 1 mg/kg DXM every day.(3)Control group: OVA and DXM was replaced with normal saline(NS).(4) selenomethionine low dose group and high- dose group:From the first 15 days OVA sensitized rats given intraperitoneal injection of selenomethionine 0.1mg/(kg.d)2.5 mg/(kg.d),total of 7 times.(3) Controlgroup: OVAã€DXM and selenomethionine was replaced with normal saline(NS). In the 24 hours after the lastchallenge, rats were killed to collect lung tissue and airway lavage fluid(BALF). Collect alveolar fluid(BALF) and lung tissue. lung tissue is made into lung biopsy specimens through dehydration, paraffin embedding.Interleukin 4(IL-4) and Interferon-γ(IFN-γ).levels in BALF supernatants were measured by elisa method. BALF cells and supernatant is separated by centrifugal separation. Intercellular adhesion molecule 1 levels of lung specimens were assessed with the use of immunohistochemical. Eachgroup of indicators data were analyzed and compared by statistical methods.Results1. Compared with the asthma group, dexamethasone group and selenomethionine low, and high dose groups in BALF IL-4 levels were significantly lower(P <0.05).Compared with control group, every group in BALF that IL- 4 levels were significantly increased(P <0.05); Between selenomethionine low and high dose groups, there is no statistically significant difference.2.Compared with the asthma group, dexamethasone group and selenomethionine low, and high dose groups in BALF IFN-γlevel were significantly increased(P <0.05); Compared with control group, every group in BALF that IFN- γ levels were significantly increased(P <0.05); Between selenomethionine low and high dose groups, there is no statistically significant difference.3. Compared with the asthma group, dexamethasone group and selenomethionine low, and high dose group the total number of cells in BALF neutrophils, eosinophils counts were significantly decreased(P <0.05),every group in BALF counts were significantly increased(P <0.05); Between selenomethionine low and high dose groups, there is no statistically significant difference.4. Compared with the asthma group, dexamethasone group and selenomethionine low and high dose lung tissue ICAM-1 expression was significantly lower(P <0.05).Between selenomethionine low and high dose groups, there is no statistically significant difference.ConclusionSelenomethionine and dexamethasone could significantly increase the level of IF N-γ in asthmatic rats, decreased the level of IL-4 level in BLAF, and the expr ession of ICAM-1 in lung tissues to reduce the expression of ICAM-1. |
