ObjectiveTo evaluate the ERagene methylation level in postmenopausal osteoporosis patients bone tissue.To explore the effect of Golden kidney bolus on ERagene expression and methylation level in rats bone tissue with bilateral ovariectomy.Methods1.By the methods of MassArray methylation,detected the ERagene methylation difference between PMOP female bone tissue and control group.2.Assess the ERagene methylation changes in rats bone tissue with bilateral ovariectomy after Golden kidney bolus treatment through MSP.3.Preliminary explore the relationship between ERagene expression and methylation level in rats bone tissue with bilateral ovariectomy in the means of Real-time PCR.Results1.ER gene was highly methylated in bone tissue of postmenopausal osteoporosis women.2.Golden kidney bolus reduced ER alpha gene methylation level in bone tissue of rats with ovariectomy.3.Methylation level of ERagene was negatively correlated with its expression in rats bone tissue with bilateral ovariectomy.Conclusion1. Methylated ER alpha gene inhibits its mRNA transcription, and play an important role in postmenopausal osteoporosis.2. Golden kidney bolus holds s a certain therapeutic effect on female rats osteoporosis with ovariectomy.
