Studies On The Chemical Constituents Of Cynachum Paniculatum

RATIONALES:Cynanchum paniculatum (Bge.) Kitag. is an Asclepiadaceous plant and indigenous to the most part of China. It is used as a Chinese folk medicine for the treatment of stomachache, rheumatoid arthritis and hectic fevers. Its chemical composition has long been reported, but because the main ingredient of the plant is C21 steroidal glycosides, limited to the separation methods and structure identification techniques, the 1960s and 1970s, due to the emergence and application of structural separation and purification of the NMR and MS techniques, these compounds to identify areas have a breakthrough. Until 1986, the characteristic ingredients of Cynanchum paniculatum was clarified as C21 steroidal glycosides by professor Bo Bridge. Recent studies show that its chemical composition includes a variety of C21 steroidal glycosides, flavonoids and a small amount of alkaloids.However, the chemical component and active component is not completely illuminated, which is not comparable to the importance of this famous drug.OBJECTIVE:To study chemical components isolated from the 95% ethanol extract of C. paniculatum, and lay the foundation for its pharmacological activity research.METHODS:The compounds were isolated by different chromatographic methods and their structures were identified by the physicochemical characters and the spectrascopic analysis.RESULTS:Thirty compounds were isolated and 25 of them were identified as cynatratoside A (1), glaucogenin C (2), glaucoside A (3), cynapanoside A (4), neocynapanogenin F (5), neocynapanogenin F 3-O-β-D-oleandropryranoside (6), carumbelloside II (7),3-methoxy-4,5-methylenedioxy-acetophenone (8), 1H-indole-3-carboxaldehyde (9), syringaldehyde (10),4-hydroxy-3,5-dimethoxyacetophenone (11),2,5-dihydroxy-4-methoxyacetophenone (12), paeonol (13), isopaeonol (14), apocyni(15),2,4-dihydroxyphenylethanone (16),2,5-dihydroxy-phenylethanone (17), cinnamic acid (18),p-methoxybenzoic acid (19), 3-hydroxyacetophenone (20),p-hydroxyacetophenone (21),3-methylphenol (22), benzoic acid (23),p-sitosterol (24), daucosterol (25)。CONCLUSION:Among them,8,9,11,21 and 22 were isolated from Cynachum genus for the first time; 10,15,17,18 and 23 were isolated from C. paniculatum for the first time.
