Object:To explore the correlation between ethelial ovarian cancer and HPV infection, especially high dangrous subtypes HPV16 and HPV35. Thus we can povide objective data for the prevention of epithelial ovarian cancer in Hunan province. Method:625 paraffin tissue specimens were enrolled in the study and divided into three groups: normal group, benigntumor group and cancerous group. DNA were extracted from the paraffin embedded ovarian tissue. HPV DNA was determined by PCR amplification. ① 450 bp fragments were amplified with PGMY09/11 primer set. ② The specimens of negative result were amplified secondly with GP5+/6+ primer set that the amplified fragments was 150 bp. ③ The high dangrous subtypes HPV16 and HPV35 were amplified form positive specimens above with specific primers. All results were subjected to statistical analysis.Results : The higher quality DNA was extracted from paraffin-embedded tissues. HPV DNA touchdown PCR and nested PCR detection system was established successfully. The HPV positive detection rate of normal control group, benigntumor group and cancerous group was 17.64%( 95%CI=12.43-22.87%), 37.37%( 95%CI=29.38-45.42%) and 63.35%(95%CI=58.95-68.65%)respectively. HPV positive detection rate was higher in the cancerous group than the other two groups, the difference has statistical significance( P<0.005). Two kinds of high-risk subtypes HPV16 and HPV35 were detected from HPV positive tissue. HPV16 positive detection rate was 53.97%(95%CI=45.78-61.8%)and has statistical significance(P<0.005)compared with normal group. HPV35 total positive detection rate is much lower than the HPV16 and there is statisticcal significance in the difference between the cancerous group and normal group(P<0.005).Conclusion: The results showed that the HPV infection rate in cancerous ovarian tissue is significantly higher than in normal control group and benign ovarian group, at the same time, the infection rate of benign ovarian group is higher than normal control group. It may highlight an association between HPV infection and epithelial ovarian cancer. It was also found that high-risk HPV subtype HPV16 and HPV35 account for large ratio of positive samples, and it is presumed that HPV16 maybe carcinogenic to ovary.
