Objective:The preliminary experimental results proved that Bauhinia Championii flavones(BCF) has the effect of anti myocardial ischemia reperfusion injury. The purpose of this experiment is to study the effects of BCF on regulating myocardial cell autophagy and apoptosis.Methods:1. SD rats were randomly divided into control, model, 3-MA(15 mg·kg-1), Z-VAD-FMK(1 mg·kg-1) and BCF(10, 20 mg·kg-1) groups(n=8). 30 min. ischemia, ischemia/ reperfusion 1h.or 3h, while the left anterior descending coronary artery of rats was established by ligating to make the myocardial ischemia reperfusion model.2. The changes of ST-segment on the Ⅱlead ECG were observed. The contents of creatine kinase-MB(CK-MB), inducible nitric oxide synthase(iNOS) and total antioxidative capacity(T-AOC) in serum of each group were determined according to the ultraviolet spectrophotometry. The protein expression of LC3-Ⅱ, Beclin-1 and mTOR was observed by western-blot.3. The protein expression of Bcl-2 was detected by western-blot in control, model, Z-VAD-FMK and BCF groups. The expression of Caspase-3 in myocardial tissue was observed by using Quantitative Real-time Polymerase Chain Reaction(FQ-PCR).Results:1. Effects of BCF on biochemical indexes in serum and the protein expression of LC3-Ⅱ, Beclin-1 and mTOR in ischemia reperfusion injury ratsCompared with the model group, BCF does-dependently improved T-AOC, decreased CK-MB and iNOS(P<0.05). Meanwhile, BCF down-regulated the protein expression of LC3-Ⅱand Beclin-1, as well as up-regulated mTOR protein expression(P<0.05). Compared with the 3-MA group, BCF could reduced the contents of CK-MB and iNOS, increased the level of T-AOC. It could raise LC3-Ⅱand Beclin-1 protein expression at 30 min. of ischemia, and decline the protein expression of LC3-Ⅱand Beclin-1 during reperfusion period. BCF could descend the protein expression of mTOR in ischemia/ reperfusion injury rats(P<0.05).2. Effects of BCF on the protein expression of Bcl-2 and the gene expression of Caspase-3 in ischemia reperfusion injury ratsCompared with the model group, BCF does-dependently improved the Bcl-2 protein expression. The up-regulated effect of Bcl-2 protein expression in BCF group was lower than Z-VAD-FMK group(P<0.05). The model group being used as a control, BCF does-dependently declined Caspase-3 gene expression. Compared with the Z-VAD-FMK group, BCF could raise Caspase-3 gene expression during the ischemia/ reperfusion period.Conclusions:BCF pretreatment could dose dependence to reduce myocardial ischemia/ reperfusion injury. Its protective mechanisms may be related to promote autophagy occurs at ischemia period on a short view, restrain the excessive expression of autophagy in reperfusion period and control myocyte apoptosis occurs at ischemia/ reperfusion in different periods.
