Objective To observe the influence of the Oxymatrine on the change of IL-17 A and IL-10 of spleen from asthmatic rats and explore its mechanism.Methods 40 healthy female clean SD rats were randomly divided into five groups: Normal control group, low concentration group, medium concentration group, high concentration group and the asthma model group, each group included 8 rats. The groups except the normal control group were intraperitoneal injection and atomization inhalation by ovalbumin, the normal control group same amount of saline. The rats were killed when The rats were nebulizated after 24 hours. The behavior of the rats in establish stage was observed. The lungs were observed by HE dyeing. The spleens were grinded to mononuclear cellsuspensio and adjusted the density to 2-3×106/ml by culture solution. The low concentration group, medium concentration group high concentration group were administered with different concentration of oxymatrine(1μg/ml, 10μg/ml, 100μg/ml) for 72 hours. The expression of IL-17 A and IL-10 were measured by ELISA, RT-PCR and Western blot.1. HE dyeing The existence of a large number of inflammatory cell infiltration in lung tissues of asthmatic rats in the model group, airway smooth muscle became think, excessive collagen deposition. Alveolar walls of Normal control group are smooth and pulmonary were not interstitial thicken. Lung tissues of Normal control group were not obvious inflammatory cell infiltrated. Airway smooth muscle was hyperplasia of uniform. 2. The influence on IL-17 A IL-17 A was measured by ELISA, RT-PCR and Western blot. The result compared with the normal control group, the levels of IL-17 A were increased significantly in asthma group, low concentration group and medium concentration group(P<0.05). The levels of IL-17 A had no significant differences in the high concentration group(P>0.05). And the low concentration group compared with the asthma group, the levels of IL-17 A had no significant(P>0.05). The concentrations of IL-17 A were(385.46±18.73) pg/ml,(382.21±22.24) pg/ml,(349.64±28.67) pg/ml,(231.67±27.65) pg/ml,(228.52±17.92) pg/ml. The IL-17 A m RNA expression was(83.28±15.72),(78.68±9.06),(56.12 ± 7.69),(32.50 ± 6.43),(29.89 ± 4.71). The IL-17 A protein expression were(49.94±5.43),(47.33±1.81),(29.64±2.74),(11.59±2.96),(10.49±1.59). 3. The influence on IL-10 IL-10 was measured by ELISA,RT-PCR and Western blot. The result compared with the normal control group, the levels of IL-10 were decreased significantly in asthma group, low Result concentration group and medium concentration group(P<0.05). The levels of IL-10 had no significant differences in the high concentration group(P>0.05). And the low concentration group compared with the asthma group, the level of IL-10 had no significant(P>0.05). The concentrations of IL-10 were(36.54±7.88) pg/ml,(37.18±6.95)pg/ml,(54.27±6.35)pg/ml,(86.82±7.69)pg/ml,(87.43±10.95) pg/ml. The IL-10 m RNA expression were(3.39±1.86),(4.01±2.60),(8.58±1.75),(25.73±2.59),(28.73±4.48). The IL-10 protein expression was(27.44±4.33),(30.75±2.52),(39.44±3.78),(80.05±4.01),(83.09±6.85).Conclusion 1. Oxymatrine can inhibit the expression and secretion of IL-17 A and IL-10 in asthmatic rat. 2. Oxymatrine can promote the expression and secretion of IL-10 in asthmatic rat.
