The Quality Evaluation For Authentic Ingredients Uncaria Leaves From Guizhou

Objective: To compare and analyze the quality of Uncaria hooked stems and leaves, and establish a Uncaria leaves Quality Standards for providing a reference for Uncaria leaves follow-up studies. Methods:(1)Measuring the contents of Total Alkaloid in Uncaria hooked stems and leaves with Acid Dye Colorimetry to find their difference.(2)To establish an HPLC method for the content of the rhynchophylline and isorhynchophylline which is in Uncaria stems and leaves. This method can be used to compare their content of representative compositions differences.(3)To establish HPLC fingerprint for comparative studying on chemical substances difference between Uncaria hooked stems and Uncaria leaves.(4) To establish a handy and feasible Uncaria leaves multiindex content program for it’s quality control Results:(1)The Total Alkaloid in the Uncaria hooked stems are similar with the Uncaria leaves. The Total Alkaloid in the leaves was relatively high in June and October. However, Uncaria hooked stems in December, January was relatively high.(2)The content of Isorhynchophylline in the Uncaria leaves are similar with the Uncaria hooked stems, but the Isorhynchophylline content in leaves is higher than Uncaria hooked stems in part of origin.(3)This established HPLC fingerprint conditions of Uncaria hooked stems and leaves is stabled, feasible and representative. Similarity of HPLC fingerprint was calculated by 2004 A version of “traditional medicine fingerprint similarity software evaluation system”, as a result, their similarity is higher than 0.80. It shows that they have similar chemical composition. The Uncaria leaves fingerprint peaks can be tracked in Uncaria hooked stems fingerprints when compare with Uncaria hooked stems commen fingerprint pattern that was established in the same condition. It descripts that Uncaria leaves and stems have a higher similarity for their inherent composition.(4)Uncaria leaves quality standards was eastablished by the multi-index content determination of typical composition, identification and check of related projects. Conclusion: The chemical compositions and representative composition content of Uncaria leaves are similar with Uncaria hooked stems. And the Uncaria leaves quality standards that was eastablished is simple, practical and easy to operate.This can be provided a scientific basis for Uncaria leaves as a new drug study.
