Objective: To optimizetion the best extraction conditions of polysaccharide from Dictyophora and develop a new type of beverages,,research of the beverage by Dictyophora enhanced the ability of learning and memory function in rats, Dictyophora polysaccharide on arsenic poisoning the improvement of the ability of learning and memory in rats Methods: Using single factor experiment,response surface method optimization Dictyophora polysaccharide extraction process; Single factor experiment, orthogonal experiment optimization Dictyophora beverage mixing process;Morris water maze experiment,the dark and platform experiment Dictyophora beverage to enhance learning and memory ability in norma lrats and Dictyophora polysaccharide on arsenic poisoning rats. the improvement of the ability of learning and memory by Elisa method to detect the rat BDNF in the hippocampus and cerebral cortex, Trk B, NMDA content, colorimetric method to detect the contents of Ach in the hippocampus, Ch AT, associates method to detect serum and brain tissue MDA production, hydroxylamine method to detect the contents of serum and brain tissue T-SOD, colorimetric method to detect the serum and brain tissue content of GSH and gsh-px. Results: The response surface method optimization Dictyophora polysaccharide extraction temperature 82 ℃, best condition for extracting time 3.1 h, liquid-to-material rate 62:1 ml/g, under the condition of the extraction, polysaccharide extraction yield was 13.016%, close to the model to predict the theoretical value 13.040%; Dictyophora drink best taste formula: Dictyophora polysaccharide 0.6%, sugar 7%, citric acid 0.9% and 0.02% sodium citrate; Protein content in drinks: 0.3%; Fat: 0.7%; The total sugar: 7.3%. Morris water maze experiment results: Dictyophora drink to shorten the normal rat escape latent period, increase the number of cross platform; Dictyophora polysaccharide can shorten the arsenic poisoning rat escape latent period, increase the number across platforms. Avoid dark experiment results: Dictyophora health drink can extend the incubation period of normal rats into the camera obscura, reduce the number of errors into the camera obscura, Dictyophora polysaccharide can extend the incubation period of arsenic poisoning rats into the camera obscura, decrease the number of errors for entering the camera obscura. Elisa test results: 4 weeks group and 8 weeks group hippocampus BDNF, TRKB content were higher than in normal group(P<0.05), 4 weeks group hippocampus NMDA levels higher than normal group(P<0.05); Hippocampus BDNF in low and middle dose group were higher than that of model group(P<0.05), low dose groupand high dose group hippocampus TRKB content is higher than that of model group(P<0.05). The high dose group content of hippocampal NMDA model group(P<0.05).Colorimetry test results: 4 weeks group and 8 weeks group hippocampus Ach energy is higher than normal group(P<0.05), 4 weeks group hippocampus Ch AT activity is higher than normal group(P<0.05); Dose group and high dose group hippocampus Ch AT activity is higher than the model group(P<0.05). Serum antioxidant indicators: the drinks group T-SOD content increased(P<0.05), group 2 weeks brain GSH activity and 8 weeks group higher than the control group, 4 weeks group, serum content of gsh-px and 8 weeks group higher than the control group(P<0.01), 4 weeks group of brain tissue gsh-px activity units than the control group; High dose group serum levels of T-SOD is higher than the model group(P<0.05), the dose group and high dose group T-SOD is higher than the model group(P<0.01), high dose group of serum MDA is lower than the model group, high dose group.brain GSH is higher than the model group(P<0.05), high dose group tissue gsh-px activity unit is higher than the model group(P<0.05). Conclusion: Best Dictyophora polysaccharides extraction condition rate is 13.016%; Modulate the Dictyophora polysaccharide health beverage taste comfortable, good flavor, has a unique fragrance, Dictyophora polysaccharide has certain health care function; Dictyophora polysaccharide health beverage of normal rats learning and memory ability has certain enhancement function; Dictyophora polysaccharide on arsenic poisoning rats learning and memory ability have certain improvement effect.
