Objective:by observing the head cavity clumps of chronic alcoholism rats learning and memory ability and the associated protein tyrosine hydroxylase(TH)expression and the effect of explore head hole clumps of learning and memory ability of chronic alcoholism rats and its possible mechanism of action,the influence of head for the clinical application of acupuncture point clumps of method for the treatment of chronic alcoholism caused by the ability of learning and memory disorders such as encephalopathy patients provide some experimental basis and theoretical basis.Methods:After the Morris water maze test 100 rats,the rats were randomly divided into control group 25,group 75 gavage.Intragastric ethanol feeding six weeks after the group survived 60(15 deaths during gavage)rats were randomly divided clump of scalp acupuncture group,model group and the donepezil group,each with 20,the four groups of rats again Morris water maze test.Cluster Scalp Acupuncture group:rats Baihui take its next open about 2mm at three acupoints,the needle 30 min,once daily for 4 consecutive weeks.Donepezil groups:donepezil orally treated with donepezil hydrochloride was dissolved in 0.5%sodium carboxy methyl cellulose solution,0.05mg/kg orally administered to rats treated daily with Cluster Scalp Acupuncture group performed the same period,continuous 4 weeks of treatment.Blank control group,model group:rats were crawling bundle deal,once a day,every 30min,a total of four weeks.After 4 weeks of treatment,using the Morris water maze test in rats with chronic alcoholism spatial learning ability.HE staining changes in cell morphology in the hippocampus of rats with chronic alcoholism.Observe the effects of chronic alcoholism model TH expression in rat hippocampus by immunohistochemical method.Spss 17.0 statistical package using experimental data analysis,P<0.05 was considered statistically significant.Results:1.Morris water maze test results:learning ability:building,four groups of rats escape latency comparison between groups,P>0.05,there was no statistically significant difference.Building 6 weeks later,the first hole clumps of group,model group,Donepezil escape latency of rats were significantly prolonged,longer than blank control group,P<0.05,the difference is statistically significant.Prompt building success.More than 4 weeks after treatment,Donepezil group,the escape latency clumps of head acupuncture point group were shortened,shorter than model group,is highly statistically significant(P<0.01);Head hole clumps and more Donepezil group escape latency,not statistically significant(P>0.05).Memory capacity:building before,the first four groups of rats III quadrant comparison between the residence time of groups,P>0.05,there was no statistically significant difference.Building 6 weeks later,the clumps of head acupuncture point group,model group,Donepezil together first III quadrant of the residence time of rats were significantly prolonged,longer than blank control group,P<0.05,the difference is statistically significant.Prompt building success.More than 4 weeks after treatment,Donepezil group,the clumps of head acupuncture point group first III quadrant of the residence time are shortened,shorter than model group,is highly statistically significant(P<0.01);Head hole clumps and more Donepezil group first III quadrant of the residence time,have no statistical significance(P>0.05).2.HE staining results:the blank control group,the cellular structure is complete,the outline is clear,nerve cell number,form distribution is normal,neatly arranged orderly,large,round,clear nucleoli nucleus,cytoplasm dyeing uniformity and color shallow,found no obvious pathological changes.Model group:visible disordered arrangement of a large number of neurons sparse,nucleus pycnosis,cell body shape is irregular,deep dyeing and uneven,unclear nucleoli smaller or display,nuclear membrane boundary is not clear,visible evident shell around the cell area.Head hole clumps of groups:visible nerve cells arranged neatly,cell morphological rules,clear,uniform cytoplasmic staining color shallow,rare mild contraction,pyramidal cells and nucleolus is clear,visible empty area.Donnelly,Donepezil together groups:nerve cells arranged disorder,form is more clear,cell morphological rules,yet the cell edge is clear,the most nerve nuclei is clear,rare pyknotic cones,slightly more than head hole clumps of group and obvious.3.TH expression results:head acupuncture point clumps,Donepezil group and model group obviously TH expression increase compared with blank control group,compared with the blank control group,P<0.01,the difference is statistically significant;Head hole clumps and Donepezil group TH expression is higher than the model group,P<0.01,differences according to have statistical significance;Head hole clumps of TH expression is higher than many Donepezil group,difference has statistical significance(P<0.01).Conclusion:1 The head acupuncture point clumps method can improve chronic alcoholism rats learning and memory ability,its curative effect and more pp qi group therapy effect.2 Plexus of scalp acupuncture can make chronic alcoholism rat hippocampus tyrosine hydroxylase(TH)increase,and better than donepezil.
