| ObjectiveTo explore the protective effects of epigallocatechingallate (EGCG) which is the representative monomer from Rhubarb tannins components on high glucose-induced oxidative-stress damage in HK-2 cells and to explore its possible mechanisms.To provide a reference for the comprehensive interpretation of therapeutic and toxicity effect.the substance basis and mechanism of traditional Chinese medicine Rhubarb on chronic renal injury.MethodsIn order to simulate the oxidative stress injury of kidney tissue caused by hyperglycemia in diabetic patients with kidney disease in vitro,we use the model of HK-2 cells cultured in high glucose and give the intervention effect of EGCG.The intracellular oxidative stress and antioxidant levels were determined by the cell toxicity〠the active oxygen content in cellsã€the content of MDAã€the glutathion in cells and the activity of SOD in cells. Evaluating the level of activation of endogenous antioxidant pathway Nrf2/ARE,through the protein and mRNA expression of Nrf2 in cells and its downstream antioxidant protein HO-1.ResultsHigh glucose can reduce the viability of HK-2.The content of intracellular reactive oxygen species (ROS) and the level of malondialdehyde (MDA) increased,the content of intracellular antioxidant enzymes system (GSH) and the viability of superoxide dismutase(SOD) decreased.While the EGCG may reduce the cell toxicity,improve the cell viability,decrease the level of ROS,reduce the degree of MDA,increse the content and viability of intracellular antioxidant enzymes and show a dose dependent manner to some extent.Meanwhile, it was shown that EGCG could activate Nrf2/ARE pathway at the molecular level,could induce the expression of Nrf2 protein〠mRNA and its downstream phase II detoxification enzyme HO-1 protein and improve the antioxidant ability of endogenous.ConclusionThe oxidative stress damage in HK-2 cells induced by high glucose can be reduced by EGCG,the mechanism may be related to the activation of the Nrf2/ARE pathway and it started the role of antioxidant defense of the organism itself.Protective effect of rhubarb on chronic renal injury may be related to the effect of tannin. |
