Aims:To establish the model of isolated rat myocardical ischemia-reperfusioninjury (Langendorff Model). To research the protective effect of PPAR-γagonistrosiglitazone on isolated rat myocardical ischemia-reperfusion injury.Methods:Forty healthy male Sprague-Dawley rats, whose weight were between250-300g,were randomly divided in four group. After establish the model of isolatedrat myocardical ischemia-reperfusion injury (Langendorff Model), the rats wereperfused with Kerbs-Henseleit buffer for15min(except the control group for30min),then the rats were perfused with Kerbs-Henseleit buffer in variousconcentration of rosiglitazone or GW9662for15min,after that,all groups werestopped perfused for15min,at last were repurfused with Kerbs-Henseleit buffer for60min and the coronary effluent were collected. Throughout the experiment, theLab-Chat7.2version software,which matchs with the isolated rat heart perfusiondevice made by the AD company in German,were used to recorded continuously thechanges of coronary flow and hemodynamics(HR、SBP、LVEDP、+dp/dt). Theapoptosis index(AI) by TUNEL staining was calculates, and the structural change ofmyocardial were observed by light microscope after HE staining. The concentrationof cardial troponin I(c-TnI) in coronary effluent by ELISA. The concentration ofGLUT-4in myocardial cells were detected by Western-Blot.Results:Compared with the control group, the hemodynamic parameters of groupA and B, while the group was worse. The concentration of cTn-I in groups from more to less was: G>C>Ra>Rb; TUNEL staining results are shown that AI in groups fromhigh to low was G>C>Ra>Rb; this result was the same as the HE staining abouthistomorphological structure. While, the concentration of GLUT-4and AMPK ingroups from more to less was: Rb>Ra>C> G.Discussion:Application of PPAR-γ agonist rosiglitazone pretreatment onmyocardial ischemia-reperfusion injury has a protective effect, The role of protectivedepends on the concentraction of rosiglitazone in Kerbs-Henseleit buffer. PPAR-γreceptor antagonist GW9662will aggravate the myocardial ischemia-reperfusioninjury.
