Objective: To investigate the effects of different volume artesunate on the expression of nuclear factor-κB(NF-κB)and tumor necrosis factor alpha( TNF-α) in liver tissues of mice with non-alcoholic fatty liver disease(NAFLD). Methods: The NAFLD mice model was established by providing twelve weeks of a high-fat diet, the normal group recevied a ordinary diet. The mice of successful model were randomly divided into model group, 15mg/kg/day artesunate treatment group(low dose group), 30mg/kg/day artesunate treatment group(middle dose group) and 60mg/kg/day artesunate treatment group(high dose group). low dose, middle dose and high dose of treatment groups were respectively injected into abdomen with artesunate. The nomal group and model group were treated with sodium chioride injection. The mice were sacrificed after twelve weeks’ treatment, serum and liver samples were collected for ALT、AST、TG、CHOL、GLU tests, NF-κB、TNF-α detections and pathomorphology anlysis. Results: Compared with the normal group, Levels of NF-κB、TNF-α、ALT、AST、TG、CHOL、GLU were increaed significantly in model group(P<0.05). Livers from the model group showed obvious histological features of inflammation and steatosis in hepatocytes. NF-κB was good positive correlated with TNF-α(P<0.01). Inflammation was positive correlated with NF-κB、TNF-α(P<0.05), and steatosis was positive with TNF-α(P<0.05),Compared to the model group, artesunate treatment led to significant decreases in the levels of NF-κB、TNF-α、ALT、AST、TG、CHOL、GLU, and pathobilogy of liver dramatically showed the repairing of inflammation and the steatosis in liver tissue. The high dose group was excellent. Conclusion: NAFLD mice have higher levels of NF-κB、TNF-α than their healthy counterparts. NF-κB、TNF-α may be positive related with inflammation and steatosis of liver tissue. Artesunate may play a role in treatment to NAFLD by reducing the expressiom of NF-κB、TNF-α.
