Objective:To investigate the effect of β-carboline alkaloid on proliferation of gastric carcinoma cell and its relationship with the expression of PTEN and ERK mRNA.Methods:The rate of SGC-7901 cells in vitro growth inhibition was determined by CCK-8 on different dose and time of β-carboline alkaloid. FCM were used to analyze cell apoptosis. The expression of PTEN and ERK mRNA were detected with RT-PCR.Results:β-carboline alkaloid could inhibit the proliferation of SGC-7901 cells significantly. After acting with β-carboline alkaloid, the expression of the PTEN mRNA was significantly higher and the expression of the ERK mRNA was significantly lower. They were time and concentration dependent(P<0.05). Conclusion:β-carboline alkaloid can inhibit the growth of SGC-7901 cells and induce apoptosis, the down-regulation of ERK mRNA and up-regulation of PTEN mRNA may be one of the mechanisms of the apoptosis.
