Objective:research the effective extract the apoptotic effect of β-carboline alkaloid on the on the gastric cancer SGC-7901 tumor-bearing mice model and its effect on the expression of the related gene Bcl-2 and Bax. Methods : establish the gastric cancer SGC-7901 tumor-bearing mice models,divide the tumor-bearing mice models into five groups by the random number table method, give high, medium and low-dose ofβ-carboline alkaloid according to a certain dose with the intervention of fluorouracil and normal saline,execute the tumor-bearing mice by cervical dislocation method after 21 days, then calculate the tumor inhibition rate of each group, take tissues from the tumor-bearing mice for relevant inspection;use the flow cytometer to detect the tumor cell apoptosis; carry out the light microscopic observation after paraffin embedding and section of the tumor tissue, HE stain and immumohistochemical stain; carry out the electron microscopy observation on the tumor cell apoptosis situations;detect the mRNA expressions of Bcl-2 and Bax by RT-PCR method to;detect the Bcl-2 and Bax protein expressions with Western blot method. Result : peganum harmala extract β-carboline alkaloid can inhibit the gastric cancer SGC-7901 tumor-bearing mice tumor proliferation and can induce the tumor cell apoptosis of gastric cancer SGC-7901 tumor-bearing mice.It is found that its apoptosis mechanism is related to lower Bcl- 2 expression and higher Bax expression and in a dose-dependent manner(P<0.05). Results:peganum harmala extract β-carboline alkaloid can inhibit the gastric cancer SGC-7901 tumor-bearing mice tumor proliferation and can induce the tumor cell apoptosis of gastric cancer SGC-7901tumor-bearing mice. It is found that its apoptosis mechanism is related to lower Bcl- 2expression and higher Bax expression and in a dose-dependent manner( P<0.05).Conclusion:peganum harmala extract β-carboline alkaloid can induce the tumor apoptosis of gastric cancer SGC-7901 tumor-bearing mice and it is speculated that its mechanism of action may be related to the lower Bcl-2 expression and higher Bax expression in the induction of tumor-bearing mice gastric cancer SGC-7901 cells by peganum harmala extract β-carboline alkaloid.
