BackgroundCancer is one of the important causes of human death, along with the ever increasing ultraviolet radiation, air pollution, radioactive chemical emissions increased abuse of cosmetics, skin cancer incidence has increased year by year, it gradually became victims Viagra important diseases of human health. The main means of treatment for skin cancer clinical surgery, radiotherapy, chemotherapy, exploring both a safe and effective drug therapy and convenient and practical means, a majority of scholars engaged in cancer prevention and treatment goals. Curcumin has antioxidant, an important role in anti-tumor, anticoagulant, anti-inflammatory, anti-atherosclerosis, cholesterol, and so, it is a natural activity of phenolic pigment extracted from turmeric rhizomes widespread in nature. The present study shows that curcumin can inhibit the growth of tumor cells, proliferation and induce apoptosis, anti-tumor mechanism has become a hot research, but whether it can inhibit the growth of skin cancer is unclear, research in this area has less.ObjectiveThrough this study of curcumin on A431 cell growth, proliferation and apoptosis, and explore curcumin against human epidermal squamous cell’carcinoma of the molecular mechanisms to provide new experimental theoretical basis for clinical treatment of skin cancer.Methods1. Application determined by MTT colorimetry to detect different concentrations of curcumin in different time treatment on A431 cell proliferation, the influence of 0,20,40, 80 umol/ml curcumin treatment respectively after 24 h,48 h,72 h, determined by MTT colorimetry method detection of curcumin on A431 cell proliferation ability.2. The research of curcumin on human skin squamous cell carcinoma A431 cells induced apoptosis, were as control group and experimental group A431 cell growth situation, morphological change; Single cell gel electrophoresis analysis of different concentrations of curcumin in different time dealing with A431 cell DNA damage; Flow cytometry detection of curcumin treatment effects on A431 cells apoptosis rate.3. Discuss turmeric in induction of human skin squamous cell carcinoma A431 cells the molecular mechanisms of apoptosis:Western blot method to detect curcumin influence on Caspase 3 A431 cells protein expression.Results1. Curcumin has obvious inhibitory effect on cutaneous squamous cell carcinoma A431 cell growth increment, and with the increase of concentration and action time of curcumin drugs increase the inhibition enhanced obviously, are dose dependent. The experimental group compared with control group, absorbance values and inhibition rate were statistically significant (P< 0.05).2. After the treatment by curcumin, some cells showed the apoptosis of morphological changes; Single cell gel electrophoresis the results showed that with the increase of drug concentration of curcumin and duration increased, cutaneous squamous cell carcinoma A431 cell DNA damage increased; Flow cytometry detection showed that with the increase of curcumin treatment concentration and action time increase of cutaneous squamous cell carcinoma A431 cells apoptosis rate increased.3. Western blot detection to cutaneous squamous cell carcinoma A431 cells of caspase-3 protein levels increased with the concentration of curcumin was varying degrees of increase than control group.ConclusionsCurcumin can inhibit proliferation of A431 cells and induce its apoptosis, after the treatment by curcumin,the DNA of A431 cell damaged and caspase 3 rising in protein expression may be involved in the process of apoptosis.
