Objective: to investigate the effect of curcumin on JNK/c-Jun signaling pathway to alleviate the damage of hippocampal neurons in early brain injury(EBI)after subarachnoid hemorrhage(SAH).Methods: Randomly choose 48 adult male SD rats were divided into four groups:(1)sham operation group(sham group),(2)SAH group,(3)low dose and(4)high dose group of curcumin(Cur);each group had 12 rats.The rats were sacrificed 3 days after subarachnoid hemorrhage model was established by thread method;Use Western blotting method to test the expression level of p-JNK,JNK,p-c-Jun,c-Jun and caspase-3,determination of caspase 3 expression by immune-histochemical method;TUNEL and Nissl staining measurement neuron damage.Results: The p-Jun ? c-Jun,p-JNK ? JNK,Caspase 3 expression increased in SAH group(p < 0.05)compared to the sham group;high and low dose group of Cur p-Jun/Jun,p-JNK/JNK,caspase-3 expression lowered compared to SAH group(p < 0.05);Cur high dose groups expressions lowered more than the low dose group(p < 0.05).TUNEL and Nissl staining in SAH group compared with sham group,Nuclear hyper-chromatism?cracking increased,Nissl bodies disappeared and cell lysis swelling increased;Cur high?low dose group compared with SAH group: the cell swelling ?Nissl body cracking decreased,especially the expression of high-dose group.conclusion: curcumin can inhibit neuronal apoptosis and protect EBI after SAH,which may join in regulating the expression of JNK/c-Jun signaling pathway. |
