Objective:Discussion SDF-1 monoclonal antibody carrying Ultrasound-targeted microbubble with the coronary artery transplantation of pig bone marrow mesenchymal stem cells (MSCs) homing relationships and explore bone marrow mesenchymal stem cell transplantation in the treatment of myocardial infarction optimal time window. Methods: The 39 Chinese mini-pigs were randomly selected 3 as a control group, according to more than 36 after myocardial infarction, Id,3d, 1w,2w,3w,4w six time points were randomly divided into six groups, each 6, each experimental group was divided into two subgroups: SDF-1 measurement group (3) and stem cell transplantation group (3). SDF-1 measurement and control groups were injected SDF-1 monoclonal antibody carrying Ultrasound-targeted microbubble (Tgroup) and common ultrasound angiography agent (C group), the measured parameters to obtain angiography agent of myocardial perfusion parameters(A, β, A×β), SDF-1 measurement set of relative quantitative PCR results,stem cell transplantation group were treated between intracoronary injection of BrdU-labeled bone marrow mesenchymal stem cells (MSCs), are on transplant 3d animals were sacrificed, the detection of myocardial infarct volume MSCs homing. Results:Targeted contrast agent in myocardial perfusion parameters(A, P, A×β)is consistent with the trend SDF-1 content trends, are the highest in one week, and then began to decline.Targeted contrast agent in myocardial perfusion parameters(A, P, A×β)and SDF-1 were correlated (r= 0.887,0.892,0.942, P<0.05), resulting in targeted contrast agent in myocardial perfusion parameters(A, β, A×β)(X) and SDF-1 (Y) of the regression equation Y= 0.699X-0.6048, Y= 0.4698X+0.3282, Y= 0.0945X+0.6685 (R2= 0.772,0.7957,0.8871, P<0.05).Targeted contrast agent in myocardial perfusion parameters (A, β, A×β)and the number of stem cells homing began to increase both from the first day, to the first seven days of the peak and then begin to decline. Targeted contrast agent in myocardial perfusion parameters A, β, and A×β and the number of stem cells homing are correlated (r= 0.658,0.778,0.777, P<0.05); β(X) and the number of stem cell homing (Y) set up a regression equation Y= 36.11+17.601X (R2= 0.605, P<0.05); A×P (X) and the number of stem cell homing (Y) set up a regression equation Y=50.023+3.348X (R2= 0.604, P <0.05).Conclusion:Targeted contrast agent in myocardial perfusion parameters values and the number of stem cell homing after myocardial infarction reached a peak at 1 week. Myocardial perfusion parameters of targeted contrast agent can be dynamically evaluated the number of stem cell homing variation of the time window, a correlation between them.
