Objective:By Langendorff heart perfusion in vitro method to establish acute myocardial ischemia model of rabbit with heart rate (HR), P-R interphase, ST segment and T wave amplitude, the left ventricular systolic pressure (LVSP), left ventricular di astolic final pressure (LVEDP) and left ventricular pressure maximum change rate (+dp/dtmax) and coronary flow and myocardial water percentage as an index, obs ervation of high, low and normal doses of acid-insoluble ash scallions Bai Guizhi decoction extract on acute myocardial ischemia heart perfusion in vitro of the rabbi t, and to explore its mechanism of action, for clinical reasonable application of aci d-insoluble ash scallions Bai Guizhi soup in treatment of acute myocardial ischemi c diseases to provide experimental basis.Methods:To establish acute myocardial ischemia model on rabbit, observation records for each process in vitro experiment heart electrocardiogram (ecg) and heart indoo r pressure curve.Determination of HR, P-R), S-T, T wave amplitude change of indicators.ST segment and T wave measurement will be subject to the baseline, v ertical distance measure each point to the baseline.Heart indoor pressure measureme nt in LVSP, LVEDP,+dp/dtmax, dp/dtmax change as an index, LVSP, LVEDP,+ dp/dtmax, dp/dtmax data, and to further study the scallions frutus aurantii immaturu s Bai Guizhi soup pretreatment on the mechanism of action of acute myocardial is chemia rabbit.Results:1. The model of rabbit electrocardiogram HR slowed significantly, P-durati on between R, S-T significantly higher, T wave amplitude, compared with the n ormal group were significant differences by statistical analysis;LVSP model group was obviously lower, LVEDP increased significantly, plus or minus dp/dtmax slowe d, compared with the normal group were significant differences by statistical analys is;Myocardial water percentage increases, model group compared with normal group were significant differences by statistical analysis;Coronary flow model group was obviously lower, compared with the normal group were significant differences by st atistical analysis.2. Acid-insoluble ash scallions Bai Guizhi decoction extract low do se group, the normal dose group HR to speed up, shorten period of P-between R, S-T period of decline, T wave amplitude is reduced, compared with model gr oup, significant differences by statistical analysis, acid-insoluble ash scallions Bai G uizhi decoction extract low dose group, normal dose LVSP recovery, reduced LVE DP,+dp/dtmax faster, compared with model group were significant differences by statistical analysis;Acid-insoluble ash scallions Bai Guizhi soup normal dose group-dp/dtmax faster, compared with model group were significant differences by statis tical analysis;Acid-insoluble ash scallions Bai Guizhi soup low dose group, normal myocardial water percentage lower dose group, compared with the normal group w ere significant differences by statistical analysis;Acid-insoluble ash scallions Bai Gui zhi soup low dose group, normal coronary flow dose group was obviously increase d, compared with the normal group were significant differences by statistical analys is.3. The scallions frutus aurantii immaturus Bai Guizhi decoction extract more tha n high dose group rabbit appeared arrhythmia, so don’t do statistical processing.Con clusion:1. By ligation of rabbit in vitro heart left anterior descending branch can b e successfully established in vitro model of acute myocardial ischemia rabbit heart. 2. The scallions frutus aurantii immaturus Bai Guizhi decoction extract low dose gr oup (0.395 g/kg), the normal dose group (0.79 g/kg) on acute myocardial ischemia model of rabbit in vitro has obvious protective effect, can promote the heart pum p function reply.3. The scallions frutus aurantii immaturus Bai Guizhi decoction ext ract high dose group (1.58 g/kg) on acute myocardial ischemia model of rabbit in vitro has a therapeutic effect, but after a period of time can cause arrhythmia.
