Objective: To observe the effect of Qilou Nephritis Decoctions on24-hour urinary protein and the levels of blood biochemical and tissuestructure of kidney and Podocin in Podocyte, investigate the functionmechanism of Qilou Nephritis Decoctions decreasing proteinuria level inADN rats.Methods: The healthy male SD rats were occasionally divided intofive groups: normal group, model group, Leigongtengduogan pian group,low-dose Qilou Nephritis Decoctions group and high-dose QilouNephritis Decoctions group. The nephrotic syndrome models of ratswere set up by injecting Adriamycin once into vena caudalis. The generalsituations of these rats were observed before and after the treatment.24-hour urine protein were measured on the second,fourth and sixthweek. After six weeks, the biochemical indices in blood serum weremeasured including blood urea nitrogen (BUN), serum creatinine (Scr),total cholesterol (TC) and albumin(Alb). And we used a light microscopyto observe the basic tissue structure of kidney and to measure theexpressions of podocin in podocyte by immunnohistochemistry staining.Results:①The24h urinary protein excretion could be reduced inthe trial groups including Leigongtengduogan pian group, low-dose QilouNephritis Decoctions group and high-dose Qilou Nephritis Decoctionsgroup.The high-dose Qilou Nephritis Decoctions group and Leigongtengduogan pian group had the most significant treatment effects.②Those trial groups could optimise the total cholesterol(TC) and albumin(Alb). The high-dose Qilou Nephritis Decoctions group andLeigongtengduogan pian group had the most significant treatment effects.③The model group of renal tubular epithelial cell degeneration,interstitial focal inflammatory cell infiltration and a large number ofprotein tube, fibrous tissue hyperplasia change was observed under a light microscopy. After the treatment with Leigongtengduogan pian or QilouNephritis Decoctions, Renal tubular epithelial cell swelling, interstitialwith a small amount of inflammatory cells and protein cast, fibrous tissuewithout proliferative changes.④The model group of Glomerularbasement membrane widened, podocyte swelling, foot processes fusionor disappeared or diffuse pathological changes was observed in atransmission electron microscrope. After the treatment with Leigongtengduogan pian or Qilou Nephritis Decoctions, We noticed glomerularbasement membrance widened slightly, podocyte swelling, partial fusionof foot process.⑤The distribution of podocin in podocyte was changed toabnormal in a varying degree in ADR rats. After the treatment withLeigongtengduogan pian or Qilou Nephritis Decoctions, the changes ofthe distribution of podocin increased.Conclusion:1. Qilou Nephritis Decoctions could reduce proteinuriaand improve serum biochemical indexes.2. Qilou Nephritis Decoctionscould improve the basic tissue structure of kidney.3. Qilou NephritisDecoctions could increase the expressions of podocin in podocyte.
