| Objective:To study theoretical basis of promoting blood circulation and dispersing phlegm in reversing PLGC; and to observe the effects of Hua-Tan-Xiao-Yu-Fang (HTXY) on expressions of Caspase-3, CyclinDl and mTOR in rats with precancerous lesions of gastric cancer (PLGC), in order to explore its pathogenesis.Methods:1. Theoretical research:Through literature collection of Traditional Chinese medicine and Western medicine, to study the cognition and treatment of PLGC, and to explore the theoretical basis and molecular mechanism that blood circulation and dispersing phlegm treat PLGC.2.100SD rats were randomly divided into the blank control group (n=17) and the model group (n=83). The rat models with PLGC were established by complex pathogenic factors, in which N-methyl-N-nitro-N-nitrosoguanidine was mainly used. After successful modeling,15rats were randomly selected from the blank control group, and75randomly selected were divided into the model group, the high dose HTXY group, the middle dose HTXY group, the low dose HTXY group, and the Vitacoenzyme group,15in each group.8weeks later, to observe the general conditions, the pathological changes of gastric mucosa, and the expressions of Caspase-3, CyclinD1and mTOR.Results: 1. In Taditional Chinese medicine, PLGC is equivalent to "distention and fullness","gastric abscess" and "Cao-za" etc. Its pathogenesis is complex, including "qi retardance and blood silts","cold coagulation" and "stagnation of phlegm","blood silts" and "stagnation of phlegm" are common pathological factors in different patients. From the angle of modern medicine, reducing phlegm and promoting blood circulation can improve the blood circulation of gastric mucosa, remove the pathogenic factor. To promote the expression of anticancer genes and to inhibit the expression of oncogenes, regulating the cell cycle, and promoting the regeneration of gastric mucosa, thereby staying the progress of the disease and preventing the gastric carcinoma. Consequently, treating PLGC by blood circulation and dispersing phlegm has significant clinical efficacy and seldom poisonous effects, it will have broad application prospects in the future.2. Compared with the blank control group, the expression of Caspase-3decreased in the model group (P<0.01), as well as the expression of CyclinD1and mTOR increased (P<0.01). Compared with the model group, the expression of Caspase-3increased in each HTXY group and the Vitacoenzyme group (P<0.01), as well as the expression of CyclinD1and mTOR decreased (P<0.01). In addition, compared with the Vitacoenzyme group, the expression of CyclinDl and mTOR decreased more significantly in the high and middle dose HTXY group (P<0.05,P<0.01).Conclusion:Hua-Tan-Xiao-Yu-Fang could obviously improve the pathological conditions of gastric mucosa in rats with PLGC. It could reverse PLGC by up-regulation the expression of Caspase-3, down-regulation the expression of CyclinDl and mTOR. |
