| objective:To investigate the pathophysiological mechanism of bcl-2 and cyclinD1 in rats with precancerous lesion of gastric cancer.By using modified Qifangweitong granules which is the Chinese medicine compound prescription with definite curative effect in clinic to intervene the rat model of precancerous lesion of gastric cancer,exploring the intervention effect of modified Qifangweitong granules" on bcl-2 and cyclinD1 protein;and to investigate the effect of "modified Qifangweitong granule" on precancerous lesion of gastric cancer;not only providing experimental evidence for the prevention and treatment of precancerous lesions of gastric cancer with "modified Qifangweitong granule",but also providing theoretical basis for its clinical promotion.Methods: 60 rats were randomly divided into four groups,which are the blank group,model group,the group of the stomach,and the seven party stomach group,and including 15 rats in each group.Blank group was given normal drinking water.The other three groups drinking straight 180 mu g / ml n-methyl-n ’-nitro-N-nitrosoguanidine liquid(MNNG)for three month.after replicating successful model,model group were given physiological saline and Weifuchun group were given aqueous solution of Weifuchun piece,and modified Qifangweitong granules group received modified Qifangweitong free decoct Chinese traditional medicine granule,1 times / day.After 30 days of continuous treatment,the expression of bcl-2 and cyclinD1 protein were observed by immunohistochemical method and Blot Western method.Results: The two proteins that the bcl-2 and cyclinD1 were detected by immunohistochemical technique.Expression of bcl-2 in the model group was significantly higher than that in the blank group(P<0.01);After the intervention in the treatment of modified Qifangweitong granules and Weifuchun,compared with model group,the positive expression of gastric mucosa in each treatment group were decreased in different degrees(P<0.05),while there was no significant difference(P>0.05)between the group of modified Qifangweitong granules and Weifuchun.In the expression of cyclin D1,the positive expression of gastric mucosa in model group was significantly higher than that in the blank group(P<0.01).After the intervention of modified Qifangweitong granules and Weifuchun,the expression of the positive expression in each treatment group was lower than that in the model group(P<0.05);while the positive expression of modified Qifangweitong granules group was lower than that of the gastric double group(P<0.05).Conclusion: Modified Qifangweitong granules can protect the gastric mucosa of rats model with PLGC and can reduce the expression of bcl-2 and cyclinD1 protein,inhibiting excessive cell proliferation and promoting normal cell apoptosis,then to reverse the atrophy of gastric mucosa in PLGC rats,and to improve the intestinal metaplasia and atypical hyperplasia,and then achieve the purpose of the prevention and control of PLGC. |
