Objective:In this paper, using orthogonal experimental design to investigate ethanol concentration,dosage of ethanol, extraction time and extraction times on the effect of rhubarb anthraquinones extraction yield, With classic external standard method to establish standard curve of rhubarb and relevant methodology investigation project, provides the calculating basis for the subsequent experiment.To determine the five anthraquinones in Rheum officinale and its extract and formulation by the method named quantitative analysis of multi-components by single marker(QAMS). By rhubarb preparations and pilot study to examine different processing of rhubarb anthraquinones content in the samples.to provide security for the quality control of rhubarb.Methods:In the process of rhubarb extraction technology experiment, by rhubarb anthraquinones extraction rate as index, using the four factors three levels orthogonal table L9(34), poor with orthogonal analysis table and the effect curve respectively rhubarb phenol, emodin, emodin methyl ether, aloe emodin and rhein extraction yield, the data for statistical analysis to optimise the best extraction process. A HPLC-DAD method was developed to determine the contents of five anthraquinones, and QAMS methods were established when the five components were used as internal referring substances, respectively. The relative correction factors (RCFs) between the five anthraquinones (aloe-emodin, rhein, emodin, chrysophanol, physcion) were calculated and used to assay the contents of five anthraquinones in different batche Rheum officinale and formulations, and compared standards’calibration curves (SCCs) method to evaluate the accurary of QAMS method. And the relative retention value method is adopted to pinpoint chromatographic peak of5kinds of component.When the pilot tests, through volumizing rhubarb preparation into rhubarb medicinal herbs, rhubarb liquid extract, rhubarb concentrate and rhubarb liquid to spray powder samples, to determine of total anthraquinone content in the sample and compare the difference in size. Results:Get the best extraction process for anthraquinones is:Ten times the amount of60%ethanol extraction, extraction3times, each time1h. When emodin as internal referring substance, the ranges of the relative error between QAMS and SCCs in different batches Rheum officinale and extrations and preparations were-3.91%~4.06%,-3.22%~3.27%,-5.39%~7.00%, respectively. The RSD of the relative retention value were3.87-9.92%, explaining the method of QAMS have high accuracy, good repeatability. Rhubarb pilot experiment, rhubarb anthraquinone extraction yield is about43%, rhubarb medicinal powder spray powder rate and test in laboratory, compared to about27%of the yield rate is not high, still pilot is21.4%.Conclusions:Available from the results, the orthogonal experimental design is a uniform comparability, and can effectively compared to other factors, So the rhubarb anthraquinone extraction process is optimized by orthogonal design method is feasible. A rapid, low-cost and convenient QAMS method can be used as a new quality assessment model for quanlity analysis of anthraquinones in Rheum officinale and formulation.Pilot, anthraquinone in rhubarb spray powder quantitative relationship shows that alcohol extraction of rhubarb anthraquinone method in the production process is feasible.
