Obesity has always associated with a cluster of metabolic syndromes, such as insulin resistance, type2diabetes (T2D), cardiovascular diseases and so on. Actually they were all highly related to chronic inflammation. Besides, obesity is accompanied with substantial changes in composition and metabolic function of gut microbiota, which is associated with the onset of diabetes and tissue inflammation. Thus, chronic inflammation and gut microbiota are emerging targets for treating or preventing obesity and other metabolic disorders.It is reported that Ampelopsis grossedentata is involved in resisting atherosclerosis, playing a significant role in down-regulating blood lipid and blood glucose, and has such functions as anti-inflammatory, bacteriostasis, etc. But it is still unknow whether it has anti-obesity activity. Based on the previous results of our lab, C57BL/6J mice were adopted to high-fat-high-sucrose diet to establish obese model. After different periods of obese induction, extract from Ampelopsis grossedentata (EX, lOg/kg) was applied to study the therapeutic effect and the probable mechanisms were also analyzed. Results showed that the application of the extract reduced not only body-weight and fat accumulation but also the concentrations of serum TQ TC and FFA to different extent.In order to investigate the mechanism of the anti-obesity activity of Ampelopsis grossedentata extract, we checked the expression of genes related to obesity, the relative ratio of Firmicutes and Bacteroidetes in gut microbiota, and the expression of genes related to chronic inflammation. Firstly, we found that the mRNA expression of HSL, PPARα and adiponectin were up-regulated to different extent by the treatment of Ampelopsis grossedentata extract, which indicated that fat mobilization and lipid catabolism were accelerated; UCP2were also up-regulated to enhance energy consumption. As the obesity getting worse, PPARy and FAS were up-regulated while HSL were down-regulated significantly by the treatment of the extract, which will contribute to the adipocyte differentiation and fat synthesis, reduce the lipid catabolism, both attenuated ectopic lipid accumulation. Secondly, Ampelopsis grossedentata extract may also improve the chronic inflammation caused by the high-fat-high-sucrose diet. The pro-inflammatory factors such as IL-6, TNF-α were all down-regulated after the treatment of the extract. Thirdly, Ampelopsis grossedentata extract may decrease the energy intake by modulating the composition of gut microbiota. But the effect depends on the dietary habit, it works better with the basic diet.In conclusion, the extract of Ampelopsis grossedentata has remarkable therapeutic effects on diet-induced obesity. It can modulate the expression of genes related to obesity and chronic inflammation, as well as the relative ratio of Firmicutes and Bacteroidetes in gut microbiota, which in all improved the metabolism. Ampelopsis grossedentata is of great applicating prospect and exploitation values to be utilized in the anti-obesity field.
