| Purpose: In order to clarify the effect and mechanism of traditional Chinese medicineintervening ventricular remodeling and treating chronic heart failure and to provide new ideasand experimental evidence of treating chronic heart failure (CHF) by Chinese traditionalmedicine, we study effects of Chinese medicine on the expression of TLR4pathway andinflammatory factors and molecular mechanisms by observing the improvement of theventricular remodeling and inflammatory factor of rats with experimental heart failure byusing Yiqi Yangxin Decoction.Methods: We adopted near-end ligation on bottom of left anterior descending coronaryartery method to establish the rat model with heart failure after myocardial infarction; takevalsartan, relatively strong heart piece as the positive control group, observe by the naked eyethe general condition of rats; Use ultrasonic to check heart structure; Immunoblotting methodto detect the expression of myocardial TLR4and NF-κB; Radiation immunity analysismethod in detection of the content of Ang â…¡ in myocardium and plasma; Enzyme-linkedimmune adsorbent to detect BNP, TNF-α, IL-6and CRP content; Immunohistochemicalstaining method to detect the expression of myocardial BNP;HE and Masson staining todetect myocardial damage.Results:1ã€Compared with the CHF model group, each group heart function of rats with CHFwere improved to different degrees, and there were significant differences (P<0.05), in whichtraditional Chinese medicine with high-dose group of heart function improvement is the best(P<0.01);2ã€Compared with the CHF model group, each group AngII, BNP, TNF-α, IL-6andCRP levels of rats with CHF decreased significantly, and there was statistically significant (P<0.05)3ã€TLR4, NF-κB proteins expression in myocardium of rats in the CHF model group wassignificantly higher than that in sham operation group (P<0.05); and the expression of TLR4,NF-κB protein expression in each group compared with the CHF model group weresignificantly decreased (P<0.05); 4ã€The myocardial tissue in each group of rats with CHF, observed by HE, Masson,immunohistochemical staining, the damage was alleviated in varying degrees.Conclusion: Yiqi Yangxin Decoction can improve the experimental rat with CHFejection fraction (EF); improve the CHF rat inflammatory state, reduce the ventricularhypertrophy, inhibit myocardial fibrosis. Reduce the content of plasma and myocardium ofrats with CHF AngII, BNP, TNF-α, IL-6and CRP, inhibiting myocardial tissue TLR4,NF-κB expression of inflammatory factors, which may explain that the Chinese medicine’simprovement on CHF has some relationship on its inhibition on RAAS system andinflammation pathway. |
