ObjectiveInfluenza is an acute respiratory infections caused by an influenza virus, it has strong infectivity, high rate and strong pathogenicity, and it is easy to change. If the crowd are lack of immunity to mutant, it will be easy to cause to popular or pandemic, which seriously influences on people’s life and the development of social economy. Therefore, prevention influenza becomes a hotspot.This study is to explore the effect and mechanism of Zhong Sheng Wan on the mice infected by different subtypes of influenza virus, to provide the scientific evidence for Zhong Sheng Wan used in anti-influenza, to provide methods reference for the study of the effective drugs to prevent and control the influenza virus, to provide a cheap and effective kind of Chinese medicine for flu patients, to meet the growing severe security crisis of the public social health.MethodsUsing H1N1, H5N1and H9N2Avian influenza virus whose concentration is15times of a mouse half lethal dose respectively to infect the mice to make viral pneumonia model, using H1N1, H5N1and H9N2Avian influenza virus whose concentration is2times of a mouse half lethal dose respectively to infect the mice to make mortality model and using H5N1Avian influenza virus whose concentration is100times of chick embryo half infectious dose to infect the egg to make mortality model. Through the statistical software SPSS11.0for one-way ANOVA to compare the mean of the group filled with Zhong Sheng Wan, the group filled with Positive Drug and the model group base on some independent variable indexes.(1) The variable indexes of Zhong Sheng Wan acting on H9N2Virus.-the lung index and its inhibition rate of viral pneumonia model, the death protection rate and the prolong survival rate of mortality model.(2) The variable indexes of Zhong Sheng Wan acting on H5N1and H1N1Virus:the lung index and its inhibition rate of viral pneumonia model, the death protection rate, the lung tissue morphology and the prolong survival rate of mortality model, the level of CD3+, CD4+and CD8+which come from the venous blood added anticoagulants from eyeball of mouse, and the level of TNF-α、 IFN-γ and IL-10which come from the serum separated from the venous blood didn’t add anticoagulants from eyeball of mouse.(3) The variable indexes of Zhong Sheng Wan acting on H5N1Virus:Using different concentrations of Zhong Sheng Wan respectively react60min and120min with the same vlume of H5N1Virus at a concentration of100EID50/0.1ml to compare the effect of the different concentrations of Zhong Sheng Wan.ResultsThe Anti-Influenza H9N2Virus effect of Zhong Sheng Wan:Compared with the virus control group, the high and medium-dose group of Zhong Sheng Wan can significantly reduce the lung index of pneumonia caused by infection influenza H9N2virus (P<0.01), the lung index of the low-dose group is lower than the virus control group (P<0.05). Every dosage group of Zhong Sheng Wan can improve the survival and prolong the lifetime of the mice infected with the virus (P>0.05).The Anti-Influenza H5N1Virus effect of Zhong Sheng Wan:Compared with the virus control group, the low dose group of Zhong Sheng Wan can significantly reduce the lung index of pneumonia caused by infection influenza H5N1virus (0.01<P<0.05), but the high dose group of Zhong Sheng Wan has a weaker effect (P>0.05). The high and low-dose group of Zhong Sheng Wan can obviously increase the levels of CD3+T-cell (P<0.01),at the same time, the high dose group can significantly improve the levels of CD3+T-cell (P<0.01), but the role of Zhong Sheng Wan is weaker than the one of (P<0.01). The high dose group of can significantly decrease the levels of TNF-α (P<0.01), and all of the high dose group, the low dose of Zhong Sheng Wan and the group of Tamiflu can improve the level of IFN-γ and IL-10(P>0.05), but Zhong Sheng Wan is better than Tamiflu (P>0.05). All of the group of Zhong Sheng Wan and Tamiflu can relieve the symptoms of the lung interstitial edema, the alveolar damage, the degeneration of the pulmonary bronchus epithelium and the injury of bronchi wall of the mice. Meanwhile,at at room temperature, Zhong Sheng Wan reacted120min with the same vlume of H5N1Virus at a concentration of100EID50/0. lml can kill H5N1Virus at a concentration of100mg/ml.The Anti-Influenza H1N1Virus effect of Zhong Sheng Wan:Compared with the virus control group, all of the group of Zhong Sheng Wan and Lian Hua Qing Wen can significantly reduce the lung index of pneumonia caused by infection influenza H1N1virus(P<0.01), and the group of Zhong Sheng Wan is more favorable than the group of Lian Hua Qing Wen. The medium-dose group of Zhong Sheng Wan can significantly improve the survival and prolong the lifetime of the mice infected with the virus (P<0.01), the other of groups can weakly improve the survival and prolong the lifetime of the mice infected with the virus (P>0.05). The state of the mice’s lung tissue is improved through taking Zhong Sheng Wan and Tamiflu, and the effect of them is nearly. The thymus index and spleen index of the groups of Zhong Sheng Wan is higher than the one of the virus control group. All of the groups of Zhong Sheng Wan can obviously increase the levels of CD4+T-cell (P<0.01). The high and middle-dose group of Zhong Sheng Wan can obviously increase the levels of CD8+T-cell (P<0.01), the low-dose group of Zhong Sheng Wan can weakly increase the levels of CD8+T-cell (P>0.05). All of the groups of Zhong Sheng Wan can increase the levels of IFN-γ (P<0.05).The high and middle-dose group of Zhong Sheng Wan and the group of Tamiflu can obviously increase the levels of IL-10(P<0.01)GonclusionThese experimental results suggest that:Zhong Sheng Wan has some anti-influenza virus effect on.H1N1、 H5N1and H9N2virus. The high dose group of Zhong Sheng Wan has a little stronger effect than Tamiflu on anti-influenza H5N1and H9N2virus, and has weaker effect on anti-influenza H1N1virus. Zhong Sheng Wan can regulate the body’s immune function through controlling the level of T-cells, reduce the damagement of immunopathogenesis caused by virus infection through increasing the level of TNF-a and IL-10, and strengthen the activity of TH1cells to help CTL cell、NK cell and macrophages to clear away the virus. Although the ablity of Zhong Sheng Wan increasing the level of CD3+ T-cell and CD4+T-cell is weaker than the one of Tamiflu, it can inhibit the Level of TNF-α to reduce the damagement of immunopathogenesis that Tamiflu can’t.
