Objective:To central com posite design response surface m ethodologyMonomeric flavonoids investigated in vitro drug combination anti-tumorand()anti-anoxic injury effects, to find the best combination of flavonoidmonomer ratio of drugs to improve the effectiveness of the drug in theclinical treatment. Methods:Uing MTT viability calculations to reflect the sample ofanti-tumor and anti-anoxia analysis, using biochemical assay forintracellular malondialdehyde (MDA) and superoxide dismutase (SOD) level ofdetection. Using Design Expert software for quadratic regression resultsestablish a mathematical model to predict. According to the binomial equationfitting factor trend indicators drawn on the impact of the three-dimensionaland two-dimensional response surface contour map, according to thethree-dimensional and two-dimensional response surface contour map andselect the best ratio of experimental verification. According to theexperimental data to calculate the deviation of each indicator to illustratethe mathematical model to predict the accuracy of the experimental resultssize. Results:In the anti-tumor experiments,Luteolin, kaempferolquercetin use of monomers with three human ovarian cancer synergisticanti-tumor effects sensitizing, predictive value (60.48%±5.87),theexperimentally measured values are(65.09%±5.28,64.61%±3.49).Three monomers combined use of anti-tumor effects on MCF-7through20star pointset of experiments designed to the highest inhibition rate was80.52%, whilethe anti-tumor effect of luteolin monomer inhibition rate was83.73%, therole of breast cancer can be seen not as luteolin on the anti-tumor effectfor breast cancer monomer combinations. In the experiment, the antioxidant, and hydrogen peroxide after12hours on the vast majority of cell death orcell has reduced activity, as compared with the group of oxidative damage, the protection group can be administered to maintain high cell viability.Three monomers have a degree of antioxidant is administered in combination, but not too strong antagonistic oxidative damage of hydrogen peroxide,Predictive value(62.79%),the experimentally measured value(67.09%±6.28,68.61%±7.49). Biochemical parameters of SOD control group (149.10±7.67),oxidative injury group (34.43±5.67), predicted group (95.34±5.53) andMDA values of the control group (1.87±4.65), oxidative injury group(3.65±4.78), forecasting Group (2.12±5.43).Conclusion:This topic adopts the experimental model is established byCentral Composite Design Response Surface Methology, flavonoid monomercombinations of drugs and using the method of three factors and five levelsof anti-tumor and anti-oxidation in experimental research, simple and easy, good predictability, accuracy. The results confirmed, Central CompositeDesign Response Surface Methology in the Traditional Chinese Medicinecomplex experimental study it has the multi factorinteraction between multilevel.Simplify the experimental procedure, for improving the experimentalaccuracy and other advantages, it is expected to provide a new scientificmethod for the modern research of Traditional Chinese Medicine.
