The Expeirmental Reseach Of Liuchongguling Capsuleson Pharmacodynamics In Animals

Objective: To verify the pharmacological effects of Liuchongguling Capsulesand reveal its effectiveness by the related pharmacological experiment,including the effect of anti-inflammatory,analgesic,promoting bloodcirculation, immunosuppression and inhibitory effectof secondary lateral plantar swelling reaction induced by adjuvant arthritisin rats. This study provides pharmacological evidence for clinical rationaldrug use. Methods:Dimethylbenzenewas used to cause swelling of earlap skin in mice,paper was used to causegranuloma in rats to observe the anti-inflammatory effect of LiuchonggulingCapsules.Glacial acetic acid and hot plate were used to induce writhing andpain and to study the analgesic effect of Liuchongguling Capsules.Serum levelof prostaglandin E2(PGE2)in the mice with pain induced by glacial acetic acidwas measured by enzyme-linked immunosorbent assay to investigate theanalgesic mechanism of Liuchongguling Capsules. The subcutaneous injectionof adrenaline hydrochloride with ice water bath was used to prepare acute bloodstasis model in rats. Detect rats blood rheology indexes, the plasma ET-1andthe NO levels to verify the effect of invigorate the circulation of blood.The subcutaneous injection of adrenaline hydrochloride with ice water bathwas used to prepare microcirculation distuebance model in mice.Throughdetection of mice ear, the liver microcirculation blood flow, verify itsimproving microcirculation function. Through the clearance index experiment,Determining hemolysin levels, E--wreath experiment, verify the effect of Liuchongguling Capsules on nonspecific immune function, humoral immunefunction, Cellular immune function in mice.Complete Freunds adjuvant wassubcutaneously injected into the left-posterior soles of rats to induceadjuvant arthritis(AA) and to observe the inhibitory effect of LiuchonggulingCapsules on the toe swelling secondary to AA. Results: LiuchonggulingCapsules showed significant inhibitory effects on earlap skin inflammationcaused by dimethybenzene, granuloma caused by paper.And showed significantinhibitory effects on pains caused by glacial acetic acid and hot plate,aswell as serum PGE2increase induced by glacial acetic acid. LiuchonggulingCapsules can improve hemorrheology in rats model of acute blood stasis,decrease the level of ET-1, increase the level of NO in plasma,and can increasemice ear, the liver microcirculation blood flow. Liuchongguling Capsulesshowed significant effects on decreasing hemolysin levels and the amount ofE—wreath,but no significant effects on clearance index. LiuchonggulingCapsules showed significant inhibitory effects on toe swelling secondarycaused by AA. Conelusion: Liuchongguling Capsules have anti-inflammatoryand analgesic effects, can invigorate the circulation of blood, haveImmunosuppressive effect.The analgesic mechanism is related to inhibition ofA synthetic. The invigorate the circulation of blood mechanism is related todecreasing the level of ET-1, increasing the level of NO in plasma. And thecapsule has a therapeutic effect on secondary lesions caused by AA in rats.
