Objecttive:Explore adiponectin(adp) and adiponectin gene single-nucleotide polymorphisms(SNP) in the role of coronary artery disease, Analysis the relevance between SNP-11377C/G sites and SNP+45T/G sites and coronary heart disease mutations coronary lesions range of correlation.Methods:Totally225patients hospitalized received coronary angiography were included during Aug2009and Jan2010,whose main artery (the left anterior descending LAD, left circumflex LCX teams, and right coronary artery RCA) stenosis at least>50%for the inner diameter stenosis after coronary angiography formed the coronary artery disease(CAD) group(118),the control group including97cases who excluded coronary heart disease after the biochemical examination、the sternum、electrocardiogram、echocardiograph、coronary angiography, and liver and kidney fuction normal patients.All the patients’follows clinic informations was recorded, such as gender, age, smoking, drinking, CAD family history, diabetes family history, triglyceride, total cholesterol, low-density lipoprotein cholesterol, high density lipoprotein cholesterol, uric acid, fasting plasma glucose, etc. Patients plasma levels of adp were measured by enzyme-linked immunosorbent assay (ELISA).Gene DNA was extracted with AxyPrep DNA reagent. The mutation of the+45T→,-11377C→G transition of adp was investigated by Polymerase Chain Reaction-restriction-Fragment Length Polymorphism PCR-RFLP, the mutation of the+45T→G、-11377C→G transition was identified with agarose gel electrophoresis. And choose to do gene sequencing. Statistic method:Analyze the data used the statistic software SPSS16.0. Chi square test were used to compare numeration data between two groups. Scale data were expressed with mean±sd, followedT test was used to compare scale data between groups. P<0.05was considered to be statistic significance.Results:CAD group adiponectin significantly lower than the control group, and acute coronary syndrome(ACS) group below stable angina group(SAP),while no difference was found between myocardial infarction(MI) group and unstable angina (UA)group,Other the other side,the level of Plasma adp with increased coronary lesions and gradually reduced.there was no obvious difference of adp SNPs+45T/G genotype distribution and allele frequency between CAD and control group,and the concentration of adp and lipid was no associated of distribution of in SNPs+45T/G each genotype. Howevre, Adp SNPs-11377C/G genotypes allele frequency distribution has significantly differences between CAD group and control group, The percentage of the CG genotype was the hithest in CAD group, while was the CC genotype in control group,we all found that G allele frequency was higher than those in the control group.but there was no signifencently difference of existenced SNPs-11377C/G loci genotype frequencies and distributions in cases with different lesions CAD group.the level of adp was distinct from the SNPs-11377C/G differenct genotype, The concentration of adp in mutation genotypes (CG and GG) were lowerr than in CC type.however,except from lipit.Conclusion:1、CAD group adiponectin significantly lower than the control group, and acute coronary syndrome(ACS) group below stable angina group(SAP), the level of Plasma adp with increased coronary lesions and gradually reduced.Clew adipone-ctin has significant role in resisting arteriosclerosis, and reflecting of the severity of coronary artery As an important index.2. Both the plasma adp level and CAD has no relation with second exon SNPs+45TT, TG, GG genotyp.so as in subgroup of CAD and coronary lesions count. T, G allele frequency had no significant difference bet-ween yin CAD group and in control.so explain that adp SNPS+45T/G gene mutation was not risk factors for CAD.3^The promoter SNPs-11377CC,CG,GG genotype had relevace with the plasma concentration of adp. The concentration of adp in mutation genotypes (CG and GG) were lowerr than in CC type. SNPs-11377genotypes、 allele frequency distribution has significantly differences between CAD group and control group. G allele frequency was higher than those in the control group. Prompt SNPs-11377was the susceptibility genes for CAD^The level of TC, TG and LDL-C in CAD goup were higher than the control group,while the HDL-C was obviously lower than the control group. Mutations type gene lipid levels were higher than those in the control group, Although there was no obvious difference by statistical analysis, But it still can’t explain the SNPs+45, SNPs-11377gene polymorphism has nothing to do with lipid metabolism disorders,maybe some patient had already taked lipid regulation medicine.
