Objective This paper studies the howplatelet-rich plasma P(RP) effects onadipose-derived stem cells’(ADSCs) number, growthrate and adipogenic andMethod The sixsamples of abdominal adipose tissue and venous blood ewre collectedfrom the same liposuction patient. Under sterile condition,adipose tissue aws etxracted toobtain primary ADSCs in stromal vascular fraction S(V)F cells;Centrifuagtion aws used toisolate PRP from the venous blood of the same liposuction patient. Cellometer analyserand AO-PI stainingaws used to count the SVFlivingcells. eHmatology analyser was usedto count the amount of platelet cells in PRP. Cells ewre divided as treatment and controlrgoups duringin vitro epxeriment and ewre counted by cellometer. The culture fluid of thetreatment groups ewre5,%10%and15%of PRP while the control rgoup contained10%eFtal Bovine Serum F(BS). At the intervals of24h,48h,72h and69h,ADSC cell growthof control and treatment groups ewre counted.Results1() Positive result is found in SVFosteoblastic and adipoegnic conversion test;third egneration ADSCs passed the Marktest and the results are CD73,CD90positive,LAH-DR and CD45neagtive;reverse verification confirmed these cells ewre ADSCs.rFom inverse reasoningit proves that ADSCs can be collected from SVF;2() Fromcentrifugation,the platelet in PRP obtained are about4times more than whole blood;3()Compared with the treatment rgoup and control,the ADSCs found at the intervals of24h,48h,72h and96h had increased in number and density. For the intervals between48h and96h,different concentrations of PRP have accelerated the rgotwh of ADCSs singificant lyand it is statistically significant P(value <0.05). Further comparison betewen these towtime intervals usingSLD method,at the72h interval,the15%PRP treatment rgoup has ahigher cell count than control group and5%PRP rgoup P(value <0.05). At the96hinterval,there is no statistical significance P(value <0.05) ecxept5%PRP treatmentrgoup.Conclusion In vitro epxeriment shown that PRP have accelerated the rgotwh of primaryADSCs.
