The Study Of The Renal Protective Effects Of Qiuzhu Capsules On Diabetic Nephropathy

Objective： To discuss the protective function of QiZhu capsules on kidney throughobserving the influence of QiZhu capsules on the related indicators of DiabeticNephropathy (DN) rats.Methods： In this study, unilateral nephrectomy and intraperitoneal injection ofstreptozotocin (STZ) approach were adopted to establish the model of diabeticnephropathy of the rat. And the rats were randomly divided into normal group, modelgroup, QiZhu capsules group and losartan potassium group. A week later, QiZhu capsulesgroup and losartan potassium group were intragastric administration with1g/kg and1.67mg/kg separately. At the end of the eighth week, it is required to observe the BUN, Scr,HbAlc, CysC and24-hoururinary protein excretion of each group. Meanwhile, the kidneypathology was observed, and immunohistochemical method was adopted to examine theTGF-β1and CTGF expression in kidney.Results：1.Impact on biochemical indicatorsModel group BUN, Scr, HbAlc, Cys-c and24h urinary protein excretion wassignificantly higher (P<0.01); Qizhu group with the above biochemical parameters of ratswas significantly lower (P<0.01), losartan group BUN, Cys-c and24h urinary protein has asimilar effect, where Scr reduce (P<0.05), no statistically significant difference inHbAlc. In addition, Qizhu group were significantly lower than losartan group (P<0.01).2.Kidney histopathology observationModel group diffuse thickening of the glomerular basement membrane, mesangialmild to moderate thickening of capillary hemangioma-like changes, tubular expansion, thepresence of renal interstitial inflammatory cells and lymphocytes; Qizhu capsule group andlosartan group have similar pathological changes in renal tissue, but a certain extentreduced compared with the model group (P<0.05).3.Immunohistochemistry was used to detect the expression of a TGF-β1and CTGFnormal mesangial cells with no or weak expression widely colored parts, theexpression of TGF-β1and CTGF in model group was significantly higher than the normalgroup (P<0.01); Qizhu capsule group glomerular epithelial cells, mesangial cells andmesangial area also has expression, but the degree of staining decreased (P<0.05); losartan group with the model group, the expression of CTGF was decreased (P<0.05), andTGF-β1was not statistically significant.Conclusion：QiZhu capsules have protective function on the kidney of DN rats, and canalso reduce the TGF-β1and CTGF protein expression in renal tissue, to improve the renalfunction and reduce renal pathological damage and slow the progress of DN.
