The study is to investigate the consistency of the expression of nervegrowth factor(NGF)、 neural cell adhesion molecule(NCAM)、 CXCchemokine ligand12(CXCL12)、 CXC chemokine receptor type4(CXCR4)gene and protein in salivary adenoid cystic carcinoma(SACC);to investigate the relationship among NGF、NCAM、CXCL12、CXCR4with the clinical pathological parameters and their relevance; thedifferences between the different expression of neural invasion pattern; tofurther study the molecular mechanism of SACC perineural invasion, toprovide a theoretical basis for control SACC perineural invasioneffectively.60paraffin-embeded SACC samples from2001.12to2012.10wereselected from The First Affiliated Hospital Of HeBei North University. Ofthe60samples,36cases of the perineural invasion group,24cases ofthe non perineural invasion group. The histological type、neural invasionpatterns、different regional nerve cells and tumor cell distribution ofSACC were observed. The samples were made into tissue chip. In situhybridization and immunohistochemistry techniques were used to detectthe expression of NGF、NCAM、CXCL12、CXCR4in the two groups, tocollect the clinical pathological parameters, and to analyze the relevancebetween index expression and sex、histological types、tumor location andvolume; to analyze the relationship between the index expression and thedifferent neural invasion patterns. All the data were analyzed by SPSS17.0software, chi-square test and spearman rank correlation wereused. The data are considered statistically significant if p<0.05.NGF mRNA、CXCL12mRNA and CXCR4mRNA were expressed inSACC tumor cells, the positive substance was located in the cytoplasmand cell membrane of tumor cells as brownish yellow or brown granules.The ratio of NGF mRNA、CXCL12mRNA、CXCR4mRNA positiveexpression in SACC perineural invasion group were more higher than thenon perineural invasion group, the difference is statistically significant (P<0.05). The expression of NGF mRNA、CXCL12mRNA and CXCR4mRNA is not associated with sex, histological types, tumor location andvolume (P>0.05). The expression of NGF mRNA is positively correlatedwith CXCL12mRNA (r=0.356, P=0.05) and CXCR4mRNA(r=0.285,P=0.027), but the expression of CXCL12mRNA is not associated withCXCR4mRNA(r=0.226, P=0.083). The statistical results shows that,there is no difference between the different neural invasion patternstainings of NGF mRNA、CXCL12mRNA and CXCR4mRNA inSACC.NGF、CXCL12and CXCR4were expressed in SACC tumor cells, thepositive substance was located in the cytoplasm and cell membrane oftumor cells as brownish yellow or brown granules. The ratio of NGF、CXCL12and CXCR4positive expression in SACC perineural invasiongroup was more higher than the non perineural invasion group, thedifference is statistically significant (P<0.05). The expression of NGF、CXCL12and CXCR4is not associated with sex, histological types,tumor location and volume (P>0.05). The expression of NGF ispositively correlated with CXCL12(r=0.279, P=0.031)and CXCR4(r=0.451, P=0.000),but the expression of CXCL12mRNA is notassociated with CXCR4(r=0.254, P=0.050). The statistical results showsthat, there is no difference between the different neural invasion patternstainings of NGF、CXCL12and CXCR4in SACC. In situ hybridization and immunohistochemistry results showed thatthere was a negative expression of NCAM in the two groups of tumorcells of SACC, and a negative expression of NCAM in part neural tissues,We speculate that the negative expression of NCAM can reduce theadhesion of tumor cells, and promote tumor cells to invasion andmetastasis. So NCAM may not directly related with the neurotropic, butindirectly participate in the invasion and metastasis of SACC withnegative expression.There is no correlation between the expression of NGF、CXCL12and CXCR4in SACC and the clinical pathological parameters. NGF、CXCL12and CXCR4all take part in the perineural invasion of SACC.Spearman displays that the expression of CXCL12and CXCR4isassociated with NGF, suggesting that the above three factors may play asynergistic role in perineural invasion in the process of SACC. The lossof the expression of NCAM may reduce the adhesion of tumor cells andnerve cells, which plays an important role in the perineural invasion ofSACC. It is very important to detect the expression of NGF、NCAM、CXCL12and CXCR4for early prediction of metastasis of SACC.
