Objection:To study monskhood and liquorice, the author conducts the investigation about the physicochemical properties and chemical and composition and pharmacologic action and analyzed the co-relationship among monskhood and liquorice decoction before and after combination. In order to analyze the effect of "increase of efficacy and decrease of toxicity" from the tiny-composition-effect", the author try to establish the analytical model of the integral mechanism of "physicochemical properties-chemical and composition-pharmacologic action". Furthermore, through illustrating the integral compatibility and effective mechanism of TCM, this thesis keens on providing a new line of thought for the research of the special effect of combination of monskhood and liquorice.The first part:To determine the change of9sorts of the main components decoctions of monskhood and liquorice after combination. The Results show that, six sorts components belonging to monskhood has reduced, and the liquiritin in liquorice has increased while Glycyrrhizic acid and glycyrrhetinic acid has reduced. To study the composition among the decoction, the author established HPLC analysis method for monskhood and liquorice decoction before and after combination. To analysis the composition deference between monskhood and liquorice decoction before and after combination, the author uses the model recognition technology to analysis the chemical composition of these4decoctions. The Results show that there are significant differences between the decoctions. Author selected12special peaks from the fingerprint of4decoctions by PLS-DA and HCA.The second part:to analyze the physicochemical properties differences between monskhood and liquorice decoction before and after combination, the author use inductivity measurer, turbid meter, viscosity meter and surface tension apparatus to measure the physicochemical properties (inductivity, salinity, turbid, viscosity, surface tension) of four kinds of decoctions; use laser diffraction to measure particle size of these four different decoctions; analyze the physicochemical properties between these decoctions. There are indeed physicochemical properties differences between these decoctions; the inductivity and salinity of monskhood and liquorice combination are less than those of other decoctions, but the particle size of combination is larger than that of other decoctions.The third part:To analysis the rats biochemical, the author uses automatic analyzer and the chemical kit to analysis change of the indexes, related to liver and kidney and heart function (BUN and CREA, CK. ALP, ALT, AST and LDH, SOD, MDA). The results showed that the toxicity of monskhood involved in rat kidney, liver, heart function, but to improve the function of the heart; liquorice has function to protect kidney, liver, heart.In order to study the heart-hurting effects of monskhood, the author uses the BL-240biological functional system to observe the heart rate, QRS extension, and arrhythmia. The results show that single monskhood decoction and the combination decoction of monskhood and liquorice have the some toxicity for heart. The fourth part:to analyz the co-relationship among monskhood and liquorice decoction before and after combination, the author selected12characteristic peak and observe the co-relationship amaong them and physicochemical properties and pharmacologic action through the grey correlation analysis method. The results suggest that8of12characteristic peaks have a close relationship with the physicochemical properties and pharmacologic action. By utilizing the reference control substance to identify five peak among.Conclusion:These analyze the integral mechanism of "physicochemical properties-chemical and composition-pharmacologic action", and identifies that "physicochemical properties-chemical and composition-pharmacologic action" indeed has close relationship in the processes of dispense. And this thesis may provide a new line of thought for the research of the special effect of dispense of monskhood and liquorice.
