Isatis indigotica is an important traditional medicine in China, is the root of chew secco plant Bahamas blue bed and isatis indigotica fort, which can clear away heat and toxic material, cool the blood detoxification. Isatis indigotica mainly used for the treatment of diseases of viral infections especially the prevention and treatment of influenza.Previously we had found the total polysaccharides(IIP-50) of Isatis indigotica exhibited significant adjuvant activity which was extracted with water at 50℃. We firstly found one of the active polysaccharides from IIP-50 that one is a a-glucan which peak molecular weight(Mp) is 3600 Da. In the thesis, we will investigate these studies as below:(1) Evaluating the quality of the total polysaccharides from the roots of Isatis indigotica which were produced in different provinces or areas.(2) Extracting other polysaccharides(IIP-100) at 100 ℃from the residues which was extracted with water at 50 ℃.(3) Isolating the IIP-100 into three sugar fractions with DEAE-cellulose column which named IIP-100-A, IIP-100-B and IIP-100-C.(4) Studying the physicochemical properties of IIP-100-A, IIP-100-B and IIP-100-C.(5) Investigating the adjuvant effect of IIP-100, IIP-100-A and IIP-100-B with HBs Ag antigen.(6) Researching the possible action mechanism of IIP-A-1 as adjuvant with H1N1antigen. After these studies, the results were shown as below:(1) Isatis indigotica of different habitats have different yield of polysaccharides, sugar content and uronic acid content, polysaccharides molecular weight distribution of different origins are similar.(2) The physicochemical properties were different between IIP-50 and IIP-100.(3) The data exhibited that IIP-100 and its fraction IIP-100-A could improve the humoral immunity in mice which were immunized with HBs Ag antigen and their adjuvant effects were as similar as alum adjuvant.(4) The mechanism of IIP-A-1 as adjuvant was studied initially. IIP-A-1 significantly promotes the proliferation of spleen T and B lymphocytes, promote spleen cells secrete IFN-γ and IL-12. Stimulate thymocyte cell secrete IL-4. IIP-A-1 can activate the MH-S cell, promote cell proliferation and TNF-a secretion. Conclusions:(1) Anhui of isatis indigotica is benefit from preparing IIP-A-1;(2) IIP-100 and IIP-100-A have the activity of adjuvants;(3) IIP-A-1 has a good role in promoting cellular immunity and humoral immunity of H1N1 influenza immunization of mice.
