Objective: Gastric cancer is one of the common gastrointestinalmalignancy. Global gastric cancer each year934,000new cases,42%(400,000) of the number of new cases of gastric cancer prevalence andmortality in our country are more than2times the world average, about every2to3minutes a Chinese man died of stomach cancer. So far, the exactpathogenesis of gastric cancer is still not very sure, with the continuousdevelopment of medical molecular research level, have made it clear thatgastric cancer occurrence, development and result in abnormal cellproliferation and apoptosis inhibition of tumor gene (ocogene) high expressionhas important relations, at the same time the high expression of oncogene andtumor recurrence and metastasis is closely related, it is also a important causeof high mortality of gastric cancer.Signal transduction and activators of transcription (signal transducers andacti-vatorsof transcription, STATs) is a downstream effector cytokines andgrowth factor receptor signaling, the transcription signal transducer andactivator of3(STAT3) is an important one in the family throughtranscriptional regulation of downstream target genes involved intumorigenesis and development.The IPA family is a class of apoptosis regulatory proteins. Found8IPAmembers: NIPA, cLAPl, cLAPZ, XIAP, TsXIAP, MLIAp, APollon and ofSurvivin in humans. Survivin as an important member of the IAP family, is byfar the strongest found apoptosis inhibitor, which can inhibit apoptosisinduced by a variety of factors, including TNF, Fas, vP16, staurosporin andvitamin K, etc. The over-expression of Survivin is suppressed to apoptosis,played an important role in the development of gastric carcinogenesis.Experimental gastric tissue, cancer tissue of STAT3and Survivin are detected in normal gastric mucosa, so as to explore the relationship betweenSTAT3and Survivin in gastric carcinogenesis, the role plays in thedevelopment and clinical pathology the relationship between thecharacteristics, which provides the basis for gastric cancer diagnosis, treatmentand clues.Methods: SP immunohistochemical method is applied to detected theexpression of STAT3and Survivin in the50cases of gastric cancer,50casesof the proximal adjacent tissues and20cases of normal large gastric mucosa,combined with the patient’s gender, age, degree of differentiation, depth ofinvasion, lymph node metastasis comprehensive analysis and otherclinicopathological factors. Using SPSS13.0statistical software for statisticaldata processing, count data applicationχ2test, P <0.05for the difference wasstatistically significant. It analyses the relationship between the expression ofboth in both gastric cancer by using Spearman rank corrdlationg which is oneof non-parameter statistics. Application of non-parametric statistics in theSpearman rank correlation of the mutual relationship between the expressionof analysis.The reagents used: rabbit anti-human STAT3polyclonal antibody waspurchased from Bioworld company working concentration of1:200, rabbitanti-human Survivin polyclonal antibody was purchased from BioworldCompany, working concentration of1:200. Goat anti-rabbit IgG antibody kitwas purchased from Beijing Zhongshan Golden Bridge Biotechnology Co.,Ltd.. DAB color kit was purchased from Beijing Zhongshan Golden BridgeBiotechnology Co., Ltd..Results:1The expression of STAT3and Survivin in gastric carcinoma, gastric canceradjacent tissues and normal gastric tissue1.1STAT3in50cases of gastric cancer tissues, adjacent tissues, the positiveexpression rate was74%(37/50),14%(7/50), a rate of11.11%(2/18)expression in18cases of normal gastric tissue. The STAT3protein positiveexpression in gastric carcinoma was significantly higher than normal gastric mucosal tissue and cancer tissue, there was a significant difference (P <0.05).The STAT3of positive expression in cancer tissue is higher than normalgastric mucosa tissue, but there is no significant difference (P>0.05).1.2The positive expression rate of Survivin in50cases of gastric cancertissues, tissue are76%(38/50),10%(5/50).18cases of normal gastric tissueexpression rate is5.56%(1/18). The positive expression rate of Survivinprotein in gastric cancer tissues is significantly higher than that of normalgastric mucosal tissue and cancer adjacent tissues, resulting in a significantdifference (P <0.05). Survivin protein in cancer tissue positive expression rateis higher than in normal gastric mucosa tissue, but no significant difference(P>0.05).2The relationship between STAT3and Survivin protein expression andclinicopathologic features in gastric cancer tissues2.1The expression of STAT3protein in gastric cancer tissue with the patient’sgender, age and the size of the tumors were not statistically significant(P>0.05), rate of the expression of STAT3in undifferentiated lower gastriccancer was88.89%(32/36), significantly higher than high, differentiation ofgastric carcinoma that the expression rate was35.71%(5/14), the differencewas statistically significant (P<0.01). The expression of STAT3in gastriccancer tissue was correlated with the invasion depth (P <0.01); Theexpression of STAT3in gastric cancer tissue was associated with lymph nodemetastasis,, lymph node metastasis group was81.25%(26/32) obviouslyhigher than that of group without lymph node metastasis was61.11%(11/18),the difference was statistically significant (P <0.05). The expression of STAT3in Ⅲ~Ⅳstage gastric cancer tissue was93.33%(28/30) was significantlyhigher than Ⅰ~Ⅱ stage gastric cancer tissue was45%(9/20), the differencewas statistically significant (P <0.05).2.2The expression of Survivin protein in gastric cancer tissue with thepatient’s gender, age and the size of the tumors were not statisticallysignificant (P>0.05). The rate of the expression of Survivin inundifferentiated lower gastric cancer was91.67%(33/36), significantly higher than high, differentiation of gastric carcinoma that the expression rate was35.71%(5/14), the difference was statistically significant (P <0.01). Theexpression of Survivin in gastric cancer tissue was correlated with the invasiondepth (P <0.01); Survivin express ion in gastric cancer tissue wasassociated with lymph node metastasis, lymph node metastasis group was93.75%(30/32), significantly higher than that of group without lymph nodemetastasis was44.44%(8/18), the difference was statistically significant (P <0.01). The Survivin expression rate of Ⅲ~Ⅳstage gastric cancer tissues was90.00%(27/30) was significantly higher than Ⅰ~Ⅱstage gastric cancertissue was55%(11/20), the difference was statistically significant (P <0.05).3STAT3and Survivin expression in gastric cancer tissues.Using the Spearman rank correlation analysis showed that the expressionof STAT3in gastric cancer tissue and Survivin were positively correlated.(rs=0.521, P <0.01)Conclusion:(1)The expression of STAT3protein in gastric cancer tissuehad a close relationship with the tumor differentiation degree, infiltrationdepth, whether there is a lymph node metastasis and TNM staging, and nocorrelation with patient’s age, gender, tumor size. The expression of STAT3protein in low, undifferentiated gastric cancer was significantly higher thanhigh, differentiation gastric cancer tissue; The expression of gastric cancer thathad already invasion of size groups was significantly higher than that had notinvading serous membrane tissue; the expression rate of the gastric cancertissue that have lymph node metastasis was obviously higher than that withoutlymph node metastasis; the expression of Ⅲ~Ⅳstage gastric cancer tissuewas significantly higher than Ⅰ~Ⅱstage gastric cancer tissue.(2)) Theexpression of Survivin protein in gastric cancer tissue had a close relationshipwith the tumor differentiation degree, infiltration depth, whether there is alymph node metastasis and TNM staging, and no correlation with patient’sage, gender, tumor size. The expression of Survivin protein in low,undifferentiated gastric cancer was significantly higher than high, differentiat-ion gastric cancer tissue; The expression of gastric cancer that had already invasion of size groups was significantly higher than that had not invadingserous membrane tissue; the expression rate of the gastric cancer tissue thathave lymph node metastasis was obviously higher than that without lymphnode metastasis; the expression of Ⅲ~Ⅳstage gastric cancer tissue wassignificantly higher than Ⅰ~Ⅱstage gastric cancer tissue.(3) Theexpression of STAT3and Survivin in gastric cancer tissue was significantlypositively correlated,, suggesting that STAT3and Survivin stomach areclosely related with the cancer occurrence, development, invasion, metastasisand prognosis, and offers a new way for the diagnosis and treatment of gastriccancer.
