| In recent years, many studies showed that the incident of malignant tumor is inrising trend year by year. Human health has been threatened badly by malignanttumor. Therefore, it is important to develop new antitumor active substances for thetreatment of malignant tumor. The occurrence and development of many kinds ofdiseases, such as malignant tumor, renal diseases, hypertension, myocardial infarction,arteriosclerosis, neurodegenerative diseases, are related to the excess of radicals. Thus,the antioxidants have wide application prospect in disease prevention and treatment.In this paper we has studied the extraction and activities of phenolic constituents fromthe strawberries. The solvent extraction of phenolic constituents from strawberry wasstudied with single-factor and orthogonal method. The antitumor and antioxidantactivities of phenolic constituents were detected. These results would providereference basis for the application and research of deep process of strawberry.1. Optimizing the extraction process for the phenolic constituents fromstrawberryTo study the optimal extraction process for the phenolic constituents fromstrawberry. The effects of time, extraction temperature, solid-liquid ratio, the ethanolsolution concentration on the content of phenolic constituents were investigated inorthogonal experiments, and further optimizes the extraction process. The optimalextract process: solid-liquid ratio1:20times volume50%ethanol; the effects of timeis3hours; the extraction temperature is70℃. Under this optimum condition, theyield of phenolic constituents was2.46%.2. Preliminary extraction and purification of phenolic constituents fromstrawberryThe phenolic constituents were extracted according to the optimal extractionprocess and purified by AB-8. The polysaccharide was eliminated by ethanol precipitation. The phenolic constituents content in the sample was73.38%with FCmethod.3. Study on the antitumor activities of the phenolic constituents from strawberryThe cytotoxic activities of the phenolic constituents from strawberry to thehuman prostate cancer cell line PC-3, the human hepatoma cell line HepG-2and thehuman breast cancer cell line MM231were detected by MTT method. The resultsshowed: the phenolic constituents had inhibiting effect on PC-3, HepG-2and MM231cells, IC50is216.0,249.2,320.2μg/mL, respectively.4. The radical-scavenging activities of the phenolic constituents from strawberryCompared with VC,we detected the effec of phenolic constituents fromstrawberry on the·DPPH, O·-2,·OH free radical scavenging. The results showed: thephenolic constituents had obvious antioxidative activities, and the higher theconcentration was, the stronger the antioxidative effect was.5. The protective and repair effects of the phenolic constituents from strawberryon the oxidative damages of cell line SH-SY5Y exposed to H2O2Human neuroblastoma SH-SY5Y cell was exposed to H2O2in vitro to establishoxidative injury model. The MTT method was applied to examine the growth ofoxidative injury model cells treated with the phenolic constituents of strawberry. Theresult showed: the phenolic constituents (Concentration<100μg/mL) had obviousprotective effect on oxidative injury.Conclusion:Under optimum conditions, the yield of phenolic constituents in the dry powderstrawberry was2.46%. After isolation and purification, the phenolic constituentscontent was73.38%. The phenolic constituents of strawberry had antitumor andantioxidant activities. |
