Clinical Significance Of Lactic Acid’s Metabolism In Glioma Diagnosis

Objective:Contrasted of the level of protein expression and RNA volume of HIF-1α in Glioma tissue and the analysis of lactic acid(Lac) in magnetic resonance spectroscopy(MRS), discussed the clinical significance of the lactic acid’s metabolism in Glioma diagnosis.Methods:Collected 30 Glioma patients who took the resection operation in our department, the WHO grading I-II were confirming in 15 of them, and the other 15 were WHO III-IV, all cases were confirmed by the pathological inspection. All patients preformed magnetic resonance imaging(MRI) and MRS before the operation. Grouping the glioma patients for two groups, people who got WHO I-II were consist of group A, which named low grade glioma(LGG), and the others in group B which belonged to high grade glioma(HGG). Used some(15 cases) injured brain tissues which were randomly collected in the patients who suffered from trauma in the same periods as control. Detected the variation of the level of HIF-1α’s protein expression and RNA volume between glioma and trauma, contrasted with the Lac volume in MRS separately, analyzed the data from the lab and the information of MRS. All data were analyzed by GraphPad Prism5.01.Results:The level of HIF-1α’s protein expression was different between Glioma and control, data of A and B were both higher than control, the P value were both less than 0.1, while B(HGG)which was almost 1.7 times of control, higher than A(LGG) which was 1.2 times of control. The level of HIF-1α’s RNA volume in A and B were higher than control, the P value were both less than 0.05, the difference was statistically significant, while B(HGG)which was almost 2.3 times of control, higher than A which was 1.8 times of control. In MRS, volume of lactic acid was more higher in lesions than in normal control, the P value were less than 0.1 and 0.05 respectively, while B(HGG)which was almost 1.7 times of control, higher than A which was 1.2 times of control.Conclusion:Contrasted the data got in the lab and the analysis of imageological diagnosis, indicating that:We could judge metabolism condition of the glioma by analyzing the volume of lactic acid which was the final product of the glycolytic pathway in MRS and evaluate the ability of growing and invasion of glioma. This approach is very significant to the prediction of the prognosis in disease.
