Objective:To investigate the inhibitory effect of selective angiotensin Ⅱ type 1 receptor antagonist on bladder cancer xenograft of nude mice.Methods:Thirty nude mice were given subcutaneous injections of EJ-M3 cells to establish the bladder cancer xenograft;then the animal models were evenly randomized into 3 groups.low-dose(10mg/kg/d),high dose(40mg/kg/d) and normal saline groups.The tumor sizes and body weights were measured when 11 day of treatment and all the mice were sacrificed on the 30th day of treatment. The microvessel density(MVD) was assessed by immunostaining of endothelial cells for CD34 and the positive incidence of vascular endothelial growth factor was measured.Results:cell strain was successfully inoculated,bladder cancer transplanted tumor model and ATIR inhibitor model were successfully established.there was significant difference between the control group and the treated groups in tumor size with P <0.05,Tumor stroma blood vessels of tumor suppression group were significantly reduced with P<0.05,Positive incidence of VEGF of tumor suppression group was significantly than it of tumor group with P<0.05.Conclusion:AT1R inhibitor can inhibit the formation of tumor stroma blood vessels obviously,lead to inhibition of the growth of tumors and maybe ATIR inhibitor inhibitor in maligmant tumor treatment plays the role of inhibition of tumor growth.
