The metal ion-BSA systems had being obtained after metal ions were added into BSA solution in order and measured with ultraviolet absorption spectrometry and tluorescence spectrometry. The results of reaction between metal ion and BSA had obtained. BSA could combine with Cu2+,Zn2+ and Ag’well, softly with Hg+, but could not do with Ca2+ and Mn2+.Fluorescence quenching phenomenon had happened when Cu2+, Ag and Hg+ reacted with BSA respectively, the volumes of Kd were 1.978×10-3,1.061×103 and 2.057×103 mol-L-1.The ultraviolet absorption spectra of metal ion-HSA systems had being obtained after metal ions were added into BSA solution in order. The ultraviolet absorption spectra of metal ion-HSA systems could be disturbed by the drug after added drug continuously into the system progressively. The result showed that Cu2+-HSA system was interfered by cefradine, but Ag+-HAS system was did weakly. Binding capacity of cefradine with HSA was stronger than that of metal ion with HSA. The binding ability of cefuroxime with HSA was also greater than these of Cu2+-HSA and Ag+-HSA.The ultraviolet absorption spectra of the drug-HSA systems had being obtained after metal ions were added into drug-HSA systems in order. The result showed that combination between cefradine and HSA was enhanced by added Cu2+while combination between cefuroxime. and HSA was weak by added Cu2+. The combination between cefradine and HSA was enhanced by added Ag+, so did the combination of cefuroxime and HSA.The fluorescence spectra of metal ion-HSA systems had being obtained after metal ions were added into HSA solution in order. The fluorescence quenching spectra of the metal ion-HSA systems were measured after drug was added into metal ion-HSA systems. The fluorescence quenching of Cu2+ acted to HSA was static quenching, the value Kd=2.443×10-4 mol-L-1,Ks=4.092×103 L-mol-1. The fluorescence quenching type of HSA after cefradine was added into metal ion-HSA systems was static quenchine. Kd=3.493×10-8 mol·L-1,Ks=2.862x 105 L-mol-1. The fluorescence quenching type of HSA after cefuroxime was added into metal ion-HSA systems was also static quenching, Kd=3.389x108 mol.L’,Ks=2.951×104 L·mol-1. The quenching action of Ag+ to HSA was also static quenching, Kr=6.360×102 mol-L-1,Ks=1.572×10-6 L·mol-1. Kd=1.387×10-5 mol-L-1 and Ks=8.871×103L-mol-1 after added cefradine. Kd=2.496×10-5mol-L-1 and Ks=4.006x104L-mol-1 after added cefuroxime.The fluorescence spectra of drug-HSA systems had being obtained after drug was added into HSA solution. The fluorescence quenching spectra of the drug-HSA systems were measured after metal ion was added into drug-HSA systems in order. The quenching type of cefradine to HSA was mainly static quenching, Kd=9.394×10-3 mol-L-1,Ks=1.065×104L-mol-1.Kd=7.584×10-3 mol-L-1 and Ks=1.319×104 L-mol-1. After added Cu2+. Kd=7.706×10-3 mol-L-1 and Ks=1.297×104L-mol-1 after added Ag+.The quenching type of cefuroxime to HSA was mainly static quenching, Kd=6.606×10-3 mol-L-1,Ks=1.514×104 L-mol-1.Kd=3.019×10-3mol-L-1 and Ks=3.312×104L-mol-1 after added Cu2+. Kd=2.172x 10-3 mol-L-1 and Ks=4.604x 104L-mol-1 after added Ag+.
