As a kind of amino acids, dencichine has the effect of stopping bleeding, antilipolytic hormones, promoting fat synthesis and also has neurotoxic effects. Dencichine was separated from the root of ginseng by researchers for many years, but there are few reports proposed that Dencichine can be separated from other parts of ginseng such as stem leaf. The stalk and leaves of panax had high yield, but low use ratio, just 5%, these are big consumptions. In this paper, a new method to determine dencichine was built; the measurement on dencichine in the stalk and leaves of Panax notoginseng from different source were investigated, and a new dencichine administration route is explored, which is significance on integrated utilization.Firstly, we build the foundation of dencichine analysis method. a reversed phase ion-pair chromatographyto test Dencichine in different age, place, part and the Processing mode of dencichine were found. The discover on the research of Dencichine content:the dencichine content of Red State is higher than WenShan Sate. The annual stem leaf is something about 2.09%, higher than other parts.the content of Dencichine from The biennial of different parts are:flower>rhizome>main root>stem leaf> fibrous root, the content of Dencichine from The triennial stem of different parts: flower>rhizome>fibrous> main root>stem leaf, Freeze drying> normal airing>Steaming hot. And Content ofdencichine is flower>rhizome> fibrous> main root>stem leaf. This test have found Dencichine in Panax stipuleanatus for the first time. moreover, the Dencichine content is higher than others.Secondly, we Optimized the Dencichine purification process. This test adopted leaf of Panax Notoginseng as material to studies the extraction,separation, and purification of Dencichine from leaf of Panax Notoginseng. In the condensation reflux extraction process, leaf of Panax Notoginseng extraction rate as the index, the orthogonal used to content the determinationby investigating five single factors in the extraction solvent,extraction time, extraction temperature and extraction material. The optimum technological conditions were determined by means of orthogonal experimental analysis method. Extraction solvent was water, extracting time was 2 h, extraction time were 3 times, extracting temperature was 50℃, extract the material ratio was 1:10. In the separation process.732 type cation exchange resin was chosen as separate packing. Gel chromatography sephdax LH-20 is Puritied. Type 732 cation exchange resin to separate the optimum process conditions of Dencichine is Enrichment concentration is 1.35 mmol/L, Adsorption after 6 hours, the adsorption rate of 96%, secondary distilled water elution to yellow,0.1% ammonia water elution, Velocity is 2 mL/mim, each tube 5 min, the eluent of ninhydrin were negative. The elution rate was 67.98%. Collection of eluent and decompression to concentrate dry weight is notoginseng coarse sample. In the purification process, we used ethanol precipitation to remove impurities and gel column to get purified. By comparison of the two kinds of gel column, Glucan gel column LH-20 was better than the CM sephadax C-25. Purity can reach 50% and productivity is 1.32%.The last, we evaluated Dencichine percutaneous absorption.In vitro percutaneous permeabilityon different penetration enhancer and penetration enhancer in different concentrations. This experiment concludes that the distribution of apparent solubility of Dencichine is 0.98mg/mL, Oil/water partition coefficients (LogP)is-1.6 through testing the effect of dencichine. Permeation effect on the mice fur is better than pig’s. The different abilities of enhancement on dencichine: 5%NMP>5% Menthol>5% PG>5% AZ. The ability on different concentrations of penetration enhancers various:5%NMP>3%NMP>1%NMP.
