Effects Of Renewed Xuefuzhuyu Decoction On The Expressions Of ICAM-1 And VCAM-1 In Atherosclerosis Rats

Objective: To observe the effect of Renewed Xuefuzhuyu Decoction on the formation of Aortic plaque and the expressions of VCAM-1 and ICAM-1 effects on AS rats, thus providing experimental basis of Renewed Xuefuzhuyu Decoction in the treatment of AS clinical application.Methods: 50 SPF Wistar male rats aged 3 months, with the average weight of 200±10 g, were randomly divided into 5 groups: the blank control group, the model group, the lipitor group, Renewed Xuefuzhuyu Decoction in low dose group(with the crude drug concentration of 0.6g/ml) and the high dose group(the crude drug concentration of 1.2g/ml). Except the blank control group, the others are used high fat combined with intraperitoneal injection of Vitamin D3 method to form AS. At the beginning of the experiment drug intervention is involved, and the dosage was given according to the body weight. After 8 weeks, measure the thickness of intima and media thoracic aortic pathology and observe the change of local lesions through a microscope. Use Immunohistochemical method to observe the expression of ICAM-1 and VCAM-1, thus giving the score.Results: From the situation of the aortic intima media thickness of rats: the high fat-fed artery intima media thickness of each group rats were higher than those of normally-fed blank group, and the aortic intima media thickness of model group increased significantly(P<0.01), which shows that the success of the model of atherosclerosis. From the positive score of ICAM-1 and VCAM-1, Compared with the blank control group, the positive score of the model group increased significantly(P<0.01); with the model group, the treatment group the positive score in varying degrees reduce(P<0.05); compared with the Lipitor group, Renewed Xuefuzhuyu Decoction high and low dose group no significant difference(P>0.05); Compared with the low dose group, the high dose group was no significant difference(P>0.05).Conclusion: Renewed Xuefuzhuyu Decoction has some effect on the prevention and treatment of atherosclerosis, which effectively reduce the aortic intima media thickness, decreased the expression of ICAM-1 and VCAM-1 and the positive score. Therefore, we speculate that Renewed Xuefuzhuyu Decoction can improve the process of atherosclerosis by inhibiting the expression of ICAM-1 and VCAM-1.
