Objective:Scleroderma is systemic sclerosis(systemic sclerosis, SSc) belongs to the Chinese medicine "skin impediment" category, traditional Chinese medicine believes that "lung advocate fur", to explore the mechanism of treatment of pulmonary worthy representative prescription "Maimendong Decoction" in the treatment of scleroderma. Modern medicine thinks that the pathogenesis of SSc are the main pathogenesis of systemic sclerosis are mainly vascular abnormalities, and abnormal collagen autoimmune abnormality three hypotheses. The pathological changes of blood vessel is the most common and most easilyaroused the concern of the anomaly, the initial stage of arterioles and microvascularabnormalities occur in systemic sclerosis. At present, many studies have shown that,control of vascular tone is regulated by neuropeptides, vascular endothelium and plateletfactor three aspects, and blood vessel in patients with systemic sclerosis early damage andplatelet activation and other factors, resulting in abnormal release of vasoactive substances, cause nerve- endothelial, control of abnormal, resulting in vasospasm. It is also one ofthe main mechanisms of Raynaud’s phenomenon. According to Takhashi,endothelial cell injury is often accompanied by endothelial cell dysfunction. Endothelial cellis the human body a large glands, can secrete and release vasoactive substances, such as cell adhesion molecule-1(VCAM-1), intercellular adhesion molecule-1(ICAM-1), P selectin(P-selectin), NO etc.. Endothelial injury, will inevitably lead to endothelialdysfunction, endocrine disorders, often expressed as a vasoconstrictor substances such as endothelin-1(ET-1) increase and vasodilator substances such as NO decreased. The VCAM-1, ICAM-1, P-selectin are mainly mediated inflammatory and immune response,endothelin-1(ET-1), NO will directly affect the vascular contraction. So by the cultured human umbilical vein endothelial cells were observed before and after treatment, VCAM-1,ICAM-1, endothelin, P selectin, NO and other indicators of change, to provide a modernmedical support for Ophiopogon Decoction in the treatment of scleroderma and TCM "lung advocate fur" theory. Methods: 60 SD rats were randomly divided into experimental group, control group, blank control group, with 20 rats in each group.Experimental group were treated with gastric perfusion of Ophiopogon Decoction(Ophiopogon Decoction 4.24g/100g/d, 1 times daily gavage, a total of one week) and Angelica four inverse soup gavage,(Angelica four inverse soup 2.44g/100g/d, 1 times daily gavage, a total of one week) and blank control group according to the experimental group dose given the same volume of normal saline by gavage for a week. 3 groups of rats 6h after the last administration of abdominal aorta blood, centrifugal Torikami Kiyo, via inactivation, sterile filter filtration, subpackage,-70 C preservation reserve. The serum containing extracted with complete medium were diluted for 5%, 10%, 20% of the serum containing low to high, group, and add another Bozhi glycopeptide were compared as the western medicine group and Chinese medicine group. Effect of MTT was used to detect serum on proliferation of endothelial cells was detected with ELISA method, the influence of drug containing serum on ET-1 secretion of endothelial cells, reducing effects of serum containing method to detect NO by the nitrate, affect the secretion of P-selectin, VCAM-1, ICAM-1 on endothelial cells with serum containing flow cytometry technology. Results:(1) Ophiopogon Decoction effect of human vascular endothelial cells 24 h 20% when the maximum inhibition rate.(2) endothelial Ophiopogon Decoction group-1 is lower than the saline group had elevated but can significantly increase NO, to a certain extent antagonized vasoconstriction effect of endothelin-1, reduce the injury of blood vessel.(3) P-selectin and VCAM-1 Ophiopogon Decoction group compared with the saline group decreased, but ICAM-1 actually increased. Conclusion: Ophiopogon decoction can reduce the expression of P-selectin and VCAM-1 in vascular endothelial cells, increase the expression of NO, a certain extent antagonized vasoconstriction effect of endothelin-1, reduce the injury of blood vessel, thus play a role in the prevention and treatment of SSc.
