Objective To investigate the expression of micro RNA(MiRNA, miR-126) and Vascular cell adhesion molecule-1(VCAM-1) in Human Umbilical Vascular Endothelial Cells(HUVEC) which are stimulated by lipopolysaccharide, and explore the possible role of miR-126 regulating VCAM-1 expression in endothelial cells’ inflammatory reaction.Methods Divided cultured HUVEC into six groups, the control group, the model group, miR-126 mimic group, miR-126 inhibitor group, A group, B group. miR-126 mimic and miR-126 inhibitor, as well as mimic control and inhibitor control were transfected into the last four groups by Lipofectamine 2000 for 24 h, after that induced by 1 μg/ml lipopolysaccharide for 8 h. mRNA of miR-126, VCAM-1 were detected by real-time PCR; The expression levels of VCAM-1 protein were detected by Western blot. Comparing the results of different groups.Results The expression of miR-126 of the model group was down-regulated(0.056 ± 0.0041) fold compared with the control group(P<0.01). The expression of VCAM-1 mRNA was up-regulated(12.50±2.55) fold compared with the control group(P<0.01), the expression of VCAM-1 protein was also up-regulated(P<0.01). After transfected with miR-126 mimic, lipopolysaccharide-induced HUVEC which compared to those transfected with mimics control, the expression of miR-126 was obviously up-regulated(51.18±2.30) fold(P<0.01), the expression levels of VCAM-1 mRNA was obviously down-regulated(0.81±0.037) fold(P<0.01), the expression levels of VCAM-1 protein was down-regulated too(P<0.01). After transfected with miR-126 inhibitor, the expression of miR-126 was obviously down-regulated(0.032±0.011) fold compared to those transfected with inhibitor control(P<0.01), the expression of VCAM-1 mRNA was up-regulated(1.42±0.10) fold(P<0.05), and the expression of VCAM-1 protein was also up- regulated(P<0.05).Conclusions Overexpression of miR-126 can down-regulate the expression of VCAM-1 at 8 h. This indicates miR-126 can depress adhesion molecule(such as VCAM-1) expression to inhibit inflammatory responses as induced by lipopolysaccharide in HUVEC.
