| AIM:To investigate the effect of QianYangYuYin(QYYY) granules in local renal tissues of Spontaneously Hypertensive Rats(SHRs) and discuss the effect of the medicated serum of QYYYG(GYGS) on Angiotensin II-induced proliferation and inflammatory reaction in Human mesangial cells(HMCs). Elaborate the influence and possible protective mechanism of QYYYG by regulating inflammation. To search for a intervented method of Traditional Chinese medicine to provide experimental basis of hypertensive renal damage.Methods:(1) In vivo:SHRs were randomly divided into model group(SHR), group of benazepril(SHR+B), group of QYYYG(SHR+Q), group of QYYYG and benazepril(SHR+Q+B),8 only each group. And choose Wister rats as control group.Rats were filled into the stomach with different medicines according to different groups, once daily for 8 weeks.The kidneys were taken out 1 day later after the last administration in rats of each group.Effects of QYYYG on protein expression of p-ERK、p-JNK、p-P38 as assessed by Western bolt.(2) In Vitro:Wistar rats were randomly divided into normal group, group of Valsartan, group of QYYYG,10 only each group.Rats were filled into the stomach with different medicines according to different groups.Then the medicated serum was prepared.HMCs were divided into 5 groups.Except the normal group, the other HMCs were induced by 10-4mol/L Angiotensin II. And each medicated serum was intervented. Western bolt was carried out to detect protein levels of p-ERK、p-JNK and p-P38.Results:(1) QYYYG suppress the proterin expression of p-ERK、p-JNK and p-P38 as assessed by Western bolt in SHR.(2) QYGS counteracts Ang Ⅱ-induced up-regulation of p-ERK、p-JNK and p-P38 in HMCs.Conclusion:(1) QYYYG may attenuate Ang Ⅱ/MAPK pathway and decrease the renal damage.(2) QYYYG inhibits the proliferation of HMCs by inhibiting the activation of Ang II /MAPK pathway. |
