ObjectiveIn recent years, breast cancer has become the first malignant tumor with which female people may come down in Word, which is threatening women’s health with the higher death rate. Its causes are not known yet. But it is believed that breast cancer is caused by multiple factors, which means gene mutation caused by a series of carcinogenic elements and genes inherited from a person’s parents leads to the breast cancer. This research bases on the detection of the expression level of CUL7 gene in breast cancer tissue, analyzes the relation between CUL7 and breast cancer, and explores the applied value of CUL7 in breast cancer’s diagnosis and targeted therapy.MethodsⅠ have chosen 100 people with primary breast cancer randomly according to the criterion, who were hospitalized in the Department of Breast Diseases of the 3rd Affiliated Hospital of Kunming Medical University from April 2013 to March 2014. Then Ⅰ use immunohistochemistry technique to test the expression level of CUL7 in these 100 cases, and analyze the level of CUL7, and correlation among CUL7 and clinical features of breast cancer (menstrual states, sizes of lumps, metastasis of axillary lymph nodes) and molecular-biological indicators (ER, PR, HER2, Ki-67, nm23, P53).Results1. Expression level: the rate of CUL7 gene’s positive expression in these 100 breast cancer tissues is 76%.2. Correlation between the expression level of CUL7 gene and clinical features of breast cancer:2.1 Correlation between CUL7 and menstrual states of patients with breast cancer:There are 45 menopause patients with breast cancer in this investigation, among which there are 34 patients with positive expression of CUL7, and the gene’s positive expression rate is 75.55%; among the other 55 premenopause women, there are 42 patients with positive expression of CUL7, and the rate reaches 76.36%. The x2 -test value of the comparison between sample rates is 19.131, with P-value being 0.009 (P<0.05), differences of both having statistical significance.2.2 Correlation between CUL7 and sizes of breast cancer’s lumpsIn≤2cm group, there are 31 patients with lumps,12 of them with CUL7 gene’s positive expression, accounting for 38.70%; in 2cm< sizes≤ 5cm group, there are 69 patients with lumps, 64 of them with CUL7 gene’s positive expression, accounting for 92.43%. According to Chi-square test, the x2-test value of the comparison is 30.451, with P-value less than 0.001 (P<0.05), differences having statistical significance. The differences between sizes of lumps and CUL7 gene’s positive expression rates show statistical significance. For instance, CUL7 gene’s positive expression rate is low when the lump is small, while with a larger lump, the expression rate is higher.3. Correlation between CUL7 gene and molecular-biological indicators of breast cancer:3.1 Correlation between CUL7 and P53:In P53 positive group of breast cancer tissue, there are 59 patients,40 of them with CUL7 gene’s positive expression, accounting for 77.96%; in P53 negative group, there are 41 patients,36 of them with CUL7 gene’s positive expression, accounting for 87.80%. According to Chi-square test, the x2-test value of two samples is 0.2377, with P-value being 0.021 (P<0.05), differences having statistical significance. There may exist negative correlation between the two.3.2 Correlation between CUL7 and ER:In ER positive group of breast cancer tissue, there are 83 patients,62 of them with CUL7 gene’s positive expression, accounting for 74.69%; in ER negative group, there are 17 patients,14 of them with CUL7 gene’s positive expression, accounting for 82.35%. According to Chi-square test, the x2-test value of two samples is 0.131, with P-value being 0.718 (P>0.05), differences having no statistical significance. There does not exist any correlation between the two.3.3 Correlation between CUL7 and PR:In PR positive group of breast cancer tissue, there are 69 patients,51 of them with CUL7 gene’s positive expression, accounting for 73.91%; in PR negative group, there are 31 patients,25 of them with CUL7 gene’s positive expression, accounting for 80.64%. According to Chi-square test, the x2-test value of two samples is 0.531, with P-value being 0.466 (P>0.05), differences having no statistical significance. There does not exist any correlation between the two.3.4 Correlation between CUL7 and HER2:In HER2 negative group of breast cancer tissue, there are 77 patients,58 of them with CUL7 gene’s positive expression, accounting for 75.32%; in HER2 positive group, there are 23 patients,18 of them with CUL7 gene’s positive expression, accounting for 78.26%. The expression level of CUL7 in HER2 positive group is higher than the one in the negative group. According to Chi-square test, the x2-test value of two samples is 0.084, with P-value being 0.772 (P>0.05), differences having no statistical significance. There does not exist any correlation between the two.3.5 Correlation between CUL7 and Ki67:In Ki67>14% positive group of breast cancer tissue, there are 78 patients,59 of them with CUL7 gene’s positive expression, accounting for 75.64%; in Ki67≤14% negative group, there are 22 patients,17 of them with CUL7 gene’s positive expression, accounting for 77.27%. According to Chi-square test, the x2-test value of two samples is 0.025, with P-value being 0.874 (P>0.05), differences having no statistical significance. There does not exist any correlation between the two.3.6 Correlation between CUL7 and molecular typing of breast cancer:In molecular typing group of breast cancer, namely Luminal-A, Luminal-B, HER2, Basal-like, there are 17 patients of Luminal-A,13 of them with CUL7 gene’s positive expression; 69 patients of Luminal-B,18 of them with CUL7 gene’s positive expression; 4 patients of HER2 positive type,1 of them with CUL7 gene’s positive expression; 10 patients of Basal-like,9 of them with CUL7 gene’s positive expression. The x2-test values for the multiple comparison among two sample rates of the CUL7 gene’s positive expression rates of all these four molecular typing are calculated carefully, with the average P-value higher than 0.05 (P>0.05), differences having no statistical significance.Conclusions1. The positive expression rate of CUL7 in breast cancer tissues is 76%.2. Correlations between expression level of CUL7 and clinical features of breast cancer are as follows: The positive expression of CUL7 is correlated with patients’ age, sizes of lumps and P53; it is higher in older patients than in younger ones with breast cancer, and higher in T2 group than in T1 with lumps; there may exist negative correlation between the positive expression of CUL7 and P53.3. Correlations between expression level of CUL7 and molecular-biological features of breast cancer are as follows: there may exist negative correlation between the positive expression of CUL7 and P53; the positive expression of CUL7 in ER positive group is higher than in the negative one, higher in PR positive group than in the negative one, and higher in Ki67 positive group. There is not any statistical difference between CUL7 and ER, PR, CerB2, Ki-67, molecular typing and so forth.4.1 conclude that CUL7 may be a gene cancer.
