IntroductionBreast cancer is the most common malignant carcinoma of women, according to the WHO statistics,500 thousand women dead due to the breast cancer, which has become the most dangerous disease to the global health of women, whose morbidity increased by years and demonstrated youth tendency.Fresh vascularization growth of the breast tumor plays an important role in the process of the breast carcinoma. Osteopontin (OPN) is the important functional protein in the celluar matrix, mediated the junction of bone tissue cell and bone matrix, participating mineralization of bone matrix and re-absorption. OPN gained much more attention as an organic response protein in formation of urination calculus, infection and immunity of body, repair and re-construction of tissue, metastasis and infiltration of tumor. Recent studies demonstrate that vascular endothelial cell growth factor (VEGF), breast tumor kinase (Brk) and activating transcription factor-4 (ATF-4) had correlation with the progress of tumor invasion and metastasis through participating in tumor vascularization. Research for now consider the middle survival time of patients who has negative expression of OPN is longer than whose positive expression, however, the relationship with breast cancer was unknown. We performed immunohistochemical method and western blot method to detect the expression of OPN, Brk, ATF-4 and VEGF in the the breast carcinoma tissue, the adjacent normal breast tissue(2cm) and the distal normal breast to investigate the mechanism and significance of OPN, Brk, ATF-4 and VEGF in the progress of breast carcinoma. MethodsThe immunohistochemical method and western blot method were used to detect the expression of OPN, Brk, ATF-4 and VEGF in the the breast carcinoma tissue, the adjacent normal breast tissue(2cm) and the distal normal breast tissue(>5cm) from 40 cases of breast cancer. Adopt SPSS13.0 covariance analysis software to carry on covariance to learn a processing, the basis different data type chooses a different covariance method. Statistical analysis was performed using the t-test and analysis of variance(SNK-q), P<0.05 was considered statistically significant.ResultThe IHC and Western blots results show that the expression of OPN, Brk, and ATF-4 increased by turns in distal normal breast tissue(>5cm), adjacent normal breast tissue(2cm) and breast carcinoma tissue.In breast carcinoma tissue the OPN,Brk,ATF-4 and VEGF protein had no correlation with age,tumor size and histological grade, increased by lymph node metastasis,clinical stage(P<0.05).In the breast carcinoma tissue OPN,Brk,ATF-4 and VEGF had correlation with each other(P<0.05),but not in the adjacent normal breast tissue(2cm) and the distal normal breast tissue(>5cm)(P>0.05).ConclusionThe expression of OPN, Brk and ATF-4 from the same signal pathway increased by turns in distal normal breast tissue(>5cm), adjacent normal breast tissue(2cm) and breast carcinoma tissue. OPN induced the adhesion and migration of endothelial cells to accelerate vascular repair through VEGF,Brk,ATF-4,had correlation with the progress of tumor invasion and metastasis through participating in tumor vascularization.
