Objective:This work was to clear whether miR-205 can be used as a high sensitivity and strong specificity miRNA marker in peripheral blood of patients with non-small cell lung cancer; analysis the relationship between the levels of miR-205 expression in peripheral blood of non-small cell lung cancer patients and clinicopathological parameters confirmed that quantitative real-time PCR to detect the expression levels of miR-205 in peripheral blood can be used to monitor tumor metastasis and recurrence, evaluate the treatment efficacy,and assess prognosis.Methods:1. In this work,we cultured YTMLC cell lines, XMLC-05 cell lines and BEAS-2B cell lines, and then compare to the level of miR-205 expression in each of the three cell lines. MiR-205 expression was defined by 2"ΔΔCT measurements。2.The 30 cases of healthy people,30 cases of lung cancer patients without distant metastases,30 cases of patients with metastatic lung cancer are divided into three groups,and then compare to the level of miR-205 expression in each of the three groups. MiR-205 expression was defined by 2-ΔΔCT measurements。3.We will calculate the sensitivity and specificity for detection of lung cancer by miR-205 and serum tumor markers CEA, CA125, CA153, CA242.4.We analysis the relationship between the expression level of miR-205 and lung cancer serum marker levels and clinical pathological features.5.We use SPSS 17.0 statistical software to data processing. The measurement data comply with the single factor analysis, grading information comply with rank sum test, correlation analysis using Pearson Chi-Square.Results:1.The level of miR-205 express differently in the YTMLC cell line, XMLC-05 cell line and BEAS-2B cell line.The miR-205 is lower expression in BEAS-2B (0.8208+0.2553)compared with in YTMLC cells (17.5072+1.9063) and XMLC-05 cells (5.7252+1.0120); and the difference was statistically significant (P<0.05).2.The level of miR-205 express differently in healthy people 1.0730 (0.5579 1.9100), without distant metastases in patients with lung cancer 3.6588 (1.6309 10.6471) and distant metastasis in patients with lung cancer 19.6324 (6.0235 43.9682),and the difference was statistically significant (P<0.05).the miR-205 was over expression in lung cancer patients.3.The sensitivity and specificity for detection of lung cancer by miR-205 was significantly higher than that by serum tumor markers CEA、CA125、CA153 and CA242.4.Analysis of the relationship between the expression levels of miR-205and clinical pathological parameters in healthy people, without distant metastases in patients with lung cancer and lung cancer patients with metastasis was found:miR-205 expression and gender, age was not statistically significant(P>0.05).5.The miR-205 over expression in lung cancer patients was related to lung cancer with distant metastasis, lymph node metastasis and elevated levels of CEA, there was statistically significance(P<0.05).6.The miR-205 over expression in lung cancer patients had no relationship with tumor marker CA125, CA153andCA242,no significance (P>0.05)Conclusions:1.The miR-205 over expression in lung cancer cell lines suggested that miR-205 plays an important role in the development of non-small cell lung cancer.2.The miR-205 is over expression in peripheral blood of lung cancer patients, especially in lung cancer patients with distant metastases; suggesting that real time PCR to detect the expression levels of miR-205 in peripheral blood can be used to monitor tumor metastasis and recurrence, evaluate the treatment efficacy, assess prognosis.
