Objective:Evaluation recombinant human granulocyte macrophage colony stimulating factor (rhGM-CSF) gel whether can promote the vascularization and epithelialization of microskin transplantation, so as to improve the success rate of biological composite film covered microskin transplantation, we want to explore a new path through the further study of biological composite films, rhGM-CSF and clinical application.Methods:30 rabbits were randomly divided into 5 groups,6 rats in each group, and full-thickness skins of them were cut off from both backsides (about 4×4cm2) and divided into the experimental group and the control group randomly.1/10 of the removed skin was cut into microskin and replanted in the wound, the experimental group covered chitosan/konjac glucomannan biological composite film painted with rhGM-CSF; The control group only covered chitosan/konjac glucomannan biological composite film, and then packaged and fixed. The two groups were unpacked respectively 1,2,3,4 and 5 weeks after the operation to observe the appearance and wound healing status, while wound tissues were taken from symmetrical parts for HE staining and immunohistochemical detection of CD31 and PCNA, In order to evaluate wound healing status.Resultsr:In the first and the second week, wound healing in experimental group was higher than that in the control group (p<0.01); in the third week, wounds in experimental group were complete healing, the control group still had a small amount of residual wounds, two group had differences (p<0.01); in the fourth and fifth week, wounds in the both of experimental and control group were complete healing, no significant differences between two groups (p> 0.05). According to the immunohistochemical detection, CD31 on behalf of microvessel density (MVD) results, in the first, the second and the third week, the experimental group was higher than that of control group, the first and the third week (p<0.01), the second week(p <0.05); In the fourth week, there was no difference between experimental group and control group (p>0.05). The fifth week without continuing determination. The positive rate of PCNA expression:in the first and the second week, the experimental group was higher than that of control group (p<0.01); In the third week, the experimental group was lower than that of control group (p<0.01); In the fourth week, there was no difference (p>0.05). The fifth week without continuing determination.Conclusions:By external use of rhGM-CSF gel, neovascularization and epithelization of wounds were promoted, the success rate of microskin transplantation covered with biological composite film was raised, wound healing was promoted.
