Objective Discuss aquaproins-4(AQP-4) on the formation of the early trauatic brain swelling expression changes and the relationship between cerebral edema and its significance.Methods Sixty healty adult male Sprague-Dawley rats,weight from 200 g to 250 g were randomly divided into two groups(the control group and the traumatic brain injury group),the control group(SO group,n=5) cut line the same as the injury group only part of the scalp do not make other processing;the trauma group(TBI group,n=25) was divided into five groups during traumatic brain injury. After the success of the building,according to the injury after 1h,2h,8h,24 h and 72 h period is divided into fives subgroups,each group 5 only.The traumatic brain injury model was estabilshed by bumpiness of free falling body according to Feeney’s.Take the brain immediately after the rats were death,The rats’ s side of brain injury and damage to the side of the brain tissue were measured by the method of wet and dry weight and HE staining to observe the changes of brain water content in different time.By adopting the method fo immunohistochemical detection of injury of brain tissue AQP-4 expression changes in each period.Results In the control group,brain tissue morphology,water content,the expression of AQP-4 had no obvious chenge;Trauma group of 1h injury,damage to the focal area increased brain tissue water content,and gradually increased with the extension fo time with injuries,24 h peak,then began to decline,72 h moisture content is higher than the control group(P<0.05);Damage to the side of the correspondin oven regional brain tissue water content also has increased accordingly,but each time the change of brain tissue water content has no significant difference(P>0.05);Damage focal area in each period and the contralateral corresponding area compared brain tissue water content,also no significant difference(P>0.05);Trauma after TBI 1h AQP-4 set of expression which began to increase, 24 h after injury AQP-4 expression reach peak,after the AQP-4 expression showed a trend of lower,72 h compared with control group still maintain a high level(P < 0.05),and its expression change rule and the changing rule of the cerebral edema same with consistency.Results show that early after brain injury,trauatic brain edema,the expression of AQP-4 and brain water content were positively close first.Conclusion ?1h after injury of rat brain edema and the expression of AQP-4 begins to rise, 24 h after injury to peakm then gradually teduced,72 h is still higher than control group;?Early in traumatic brain injury in rats,AQP-4 expression in brain tissue and the formation and development of brain edema and changes were positively correlated, prompt AQP-4 in rats play an important role in the development of traumatic brain edema.
