Objective:To explore the bone marrow mesenchymal stem cells (BMSCs) transform into neural cells by green fluorescent protein (GFP) combined with glial cell line-derived neurotrophic factor(GDNF) gene recombinant by liposome and to promote peripheral nerve regeneration.Methods:Expansion rat bone marrow mesenchymal stem cells In vitro, select the third generation of cells, Use green fluorescent protein (GFP) combined with glial cell line-derived neurotrophic factor(GDNF) gene to induce the bone marrow mesenchymal stem cells (BMSCs) turn into neural cells. Cell morphology was continuously observed by inverted phase contrast microscope. Immunofluorescence method validation and identification of neuron specific enolization enzyme (NSE) expression.Rat sciatic nerve injury model was established and grouped and then inject the induced bone marrow mesenchymal stem cells into the sciatic nerve injury regeneration chamber model.① After 4 and 8 weeks observe the index of sciatic nerve and gastrocnemius wet quality recovery rate. ② Postoperative 8 weeks measured regeneration nerve fiber diameter, the thickness of myelin sheath and axon myelinated nerve fiber counting. ③ After 4 weeks gastrocnemius muscle cell line DAPI staining.④ Postoperative 8 weeks sciatic nerve immunohistochemical staining and use scanning electron microscope observe the neurofilament regeneration.Results:Through all of the data we can see that the the group induced by GDNF gene better than the control group, the bone marrow mesenchymal stem cells grow transfered by the glial cell line-derived neurotrophic factor gene grow activity and differentiate obviously, Immunofluorescence method validation and identification of neuron specific enolization enzyme (NSE) visible specific nerve cells, Experimental sciatic nerve were observed under scanning electron microscopy nerve regeneration is more obvious, Target muscle atrophy index is lower.Conclusion:BMSCs induced by GDNF gene differentiate quickly and can be converted into nerve cells in vitro cultivation, Specifically expressed nerve cells marker protein neuron specific enolization enzyme (NSE)。After the transfection of cells injected into rat sciatic nerve injury regeneration chamber model we can see that the indicators of the experimental group are better than the control group.
