Objective: To observe the Zedoary Turmeric Oil Injection on human bladder cancer T24 cell proliferation and vascular endothelial cell factor(VEGF) expression, comprehensive analysis and evaluation of the effect of prevention and treatment Zedoary Turmeric Oil Injection of bladder carcinoma, and provide experimental basis for clinical application and Zedoary Turmeric Oil Injection treatment of bladder tumor.Methods: different concentrations of the drug in experimental group(100, 200, 400 g/mL Zedoary Turmeric Oil Injection), drug group(100 mu g/mL hydroxycamptothecin injection) and blank control group, select the human bladder cancer cell line T24 were cultured in vitro, MTT method at different concentrations were detected in T24 cells(24h, 48 H, 72h) to inhibit the expression of VEGF cell proliferation inhibition rate and the rate of. Using SPSS 17 statistical analysis software to analyze and process the above data. Od and the inhibition rate of drugs on growth of tumor cell using average ± standard deviation, inter group comparison using t test.The inhibition rate of count data using percentage(%) said, the groups were compared using the chi square test. The data processing with 95% confidence interval, and a =0.05 and a =0.01 for the standard test, when P <0.05, indicate significant difference; when the P <0.01, said the extremely remarkable difference.Results:(1) 100 g/mL hydroxycamptothecin injection in treatment of cells, with the prolongation of time, the absorbance value decreased obviously, and the value has significant difference compared with the blank control group(P <0.01), the effect of zedoary turmeric oil injection of 100~400 cells after g/mL concentration, increases with the prolongation of time and Zedoary Turmeric Oil Injection concentration, the absorbance of T24 cells decreased gradually, with obvious time effect and dose effect, in which, 100 mu g/mL of zedoary turmeric oil injection effect of 48 h, 72 h significantly decreased the absorbance(P <0.05) cells; 200 g /mL injection effect of zedoary turmeric oil 24 h, significantly reduce the degree of absorbance(P <0.05), and effect of 48 h, 72 h absorbance decreased significantly(P <0.01); 400 g/mL Zedoary Turmeric Oil Injection respectively 24 h, 48 h and 72 h could significantly reduce its absorbance(P <0.01).(2) 100 g/mL hydroxycamptothecin injection in treatment of cells, with the extension of the time of drug action, enhance its inhibitory effect on cells, the inhibition rate was increased, the inhibition rates were 39.81%, 49.19%, 59.01%. Zedoary Turmeric Oil Injection after treating cells with the increase, and the drug concentration time, the inhibitory effect of zedoary turmeric oil on tumor cells also increased, the inhibition rate increased, and the high concentration of inhibition effect on cells close to the control of drugs. The role of drugs after 24 h, the inhibition rate of 13.21%, 24.15%, 35.61%; drug effect after 48 h, the inhibition rate were 28.22%, 38.19%, 46.97%; drug effect after 72 h, the inhibition rates were 28.27%, 42.54%, 52.52%. The statistical results show that the statistical significance between each drug group and blank control group differences(P <0.01).(3) 100 g/mL hydroxy camptothecin treatment significantly reduced the concentration of VEGF, compared with the blank control group were significant differences(P < 0.01), and the inhibition was enhanced with the extension of time and trend. After that, with the increase of Zedoary Turmeric Oil Injection concentration and time, the concentration of VEGF decreased, showed a significant time- and dosage dependence. Among them, 72 h VEGF concentration decreased significantly 100 g/mL Zedoary Turmeric Oil Injection after treatment(P <0.05), 200 g/mL Zedoary Turmeric Oil Injection after treatment of 24 h, 48 h, 72 h, VEGF concentrations were significantly decreased(P <0.05), and 400 g/mL Zedoary Turmeric Oil Injection after treatment of 24 h, 48 h, 72 h, VEGF concentrations were significantly reduce(P <0.01). And the high concentration(400 g/mL) inhibited Zedoary Turmeric Oil Injection to VEGF close to control drug hydroxycamptothecin.(4) 100 g/mL hydroxycamptothecin injection and different concentrations of Zedoary Turmeric Oil Injection treatment, inhibition of VEGF expression above the trend, and a significant time and dose dependence. Hydroxy camptothecin treatment on VRGF inhibition rate of 24, 48, 72 hours respectively 8.9%, 11.6%, 14.9%, data display with the time prolonging its inhibition also enhanced. Different concentrations of Zedoary Turmeric Oil Injection effect after 24 h, the VEGF inhibition rate were 2.7%, 5.7%, 7.2%, the effect of 48 h after inhibition rates were 5.6%, 8.1%, 9.4%, the effect of 72 h after inhibition rates were 7.5%, 9%, 12.5%. The statistical results show that the statistical significance between each drug group and blank control group differences(P <0.05). Conclusion: the study showed that Zedoary Turmeric Oil Injection has inhibitory effect on the proliferation of T24 cells, inhibits T24 cell VEGF expression, the inhibition was dose-dependent; T24 cells are zedoary turmeric oil sensitive cell line. In vitro experiments suggest that Zedoary Turmeric Oil Injection is natural plant medicine for effective prevention and treatment of bladder cancer, and the effect was concentration dependent.
